摘  要:目的分析伊丽莎白菌在白纹伊蚊不同发育阶段、组织及现场种群中的分布,并研究其对蚊卵孵化以及幼虫生长的影响。方法从实验室白纹伊蚊中分离伊丽莎白菌,通过扫描电镜、理化反应以及进化树分析,综合鉴定菌属。绝对定量法检测伊丽莎白菌在白纹伊蚊不同发育阶段、组织以及现场种群的分布情况。水中添加该细菌,分为对照组、抗生素组、白纹伊蚊-3(AdeAl-3)组和抗生素+AdeAl-3组,观察不同组别蚊卵孵化率、幼虫生存率及化蛹率的变化。结果从不同发育阶段白纹伊蚊体内分离一株细菌AdeAl-3,鉴定为米尔伊丽莎白菌。AdeAl-3在白纹伊蚊蛹和幼虫含量丰富,在成蚊中较少,不同发育阶段含量差异有统计学意义(F=18.11,P<0.05)。AdeAl-3主要分布于白纹伊蚊肠中,在唾液腺和卵巢均有分布但含量较少,不同组织间的含量差异有统计学意义(F=588.70,P<0.05)。现场白纹伊蚊种群中存在AdeAl-3。AdeAl-3组蚊卵孵化率、幼虫生存率与对照组差异均无统计学意义(χ^(2)=0.06、0.01,P均>0.05);对照组、AdeAl-3组和抗生素+AdeAl-3组这三组间的幼虫化蛹率差异有统计学意义(χ^(2)=7.22,P<0.05)。AdeAl-3组幼虫化蛹率高于对照组,差异有统计学意义(χ^(2)=5.62,P<0.05)。结论伊丽莎白菌株AdeAl-3在白纹伊蚊种群中普遍存在,对蚊卵孵化、幼虫生存无显著影响,可明显提高幼虫化蛹率,具有作为工程细菌的潜在价值。Objective To analyze the distribution of Elizabethkingia in different development stages,tissues and field populations of Aedes albopictus,and study its effects on the hatching of eggs and larval growth of Aedes albopictus.Methods The bacteria were obtained from the laboratory raised Aedes albopictus.The genus was comprehensively identified by scanning electron microscopy,physicochemical reactions and phylochemical tree analysis.The distribution of Elizabethkingia in different developmental stages,tissues and field populations of Aedes albopictus was detected by absolute quantitative method.The bacteria were divided into control group,antibiotic group,AdeAl⁃3 group and antibiotic+AdeAl⁃3 group,and the changes of egg hatching rate,larval survival rate and pupation rate in different groups were observed.Results A strain of bacterium AdeAl⁃3 was isolated from Aedes albopictus at different developmental stages,and identified as Elizabethkingia miricola.AdeAl⁃3 was abundant in pupae and larvae of Aedes albopictus,less abundant in adult mosquitoes(F=18.11,P<0.05).AdeAl⁃3 was mainly distributed in the midguts of Aedes albopictus,and salivary glands and ovaries were also detected but the content was less(F=588.70,P<0.05).AdeAl⁃3 was present in the Aedes albopictus field population.Compared with the control group,AdeAl⁃3 group had no significant effects on the hatching rate of mosquito eggs and survival ratio of larvae(χ^(2)=0.06,0.01;all P>0.05).There were significant differences in larval pupation rates in the control group,AdeAl⁃3 group and antibiotic+AdeAl⁃3 group.The pupation rate of larvae in AdeAl⁃3 group was increased compared with that of the control group,the difference was statistically significant(χ^(2)=5.62,P<0.05).Conclusion The AdeAl⁃3 was a bacterium that was ubiquitous in Ae.albopictus population,and had no significant effects on the hatching and larvae survival,but could increase the pupation rate of larvae,suggesting it might be suitable for engineering bacterial modification.
