摘 要:目的探讨霍山石斛对腹泻型肠易激综合征(diarrhea predominant-irritable bowel syndrome,IBS-D)小鼠肠道菌群的影响,为霍山石斛对IBS-D的疗效机制研究提供依据。方法将24只SPF级雄性小鼠随机分为正常组、模型组、霍山石斛组、匹维溴铵组,每组6只。采用番泻叶灌胃结合束缚夹尾应激的方法制备IBS-D模型。造模成功后,霍山石斛组和匹维溴铵组分别给予相应药物剂量灌胃,持续1周。比较各组小鼠一般情况、粪便性状、粪便含水率、腹部撤退反射评分以及采用16S rRNA基因高通量测序检测小鼠肠道菌群种类和丰度的变化。结果与正常组相比,IBS-D模型小鼠精神欠佳,排便量增多,粪便含水率和内脏敏感度均升高,肠道菌群多样性和丰度有所下降;经霍山石斛水提物干预后,与模型组相比,霍山石斛组小鼠的精神状态有所改善,粪便性状恢复正常、粪便含水率和内脏敏感度均降低。物种相对丰度和组成结果表明霍山石斛水提物增加了IBS-D小鼠肠道菌群的多样性,并且显著增加了norank_Muribaculaceae、乳杆菌属、拟普雷沃菌属等肠道有益菌属的相对丰度,降低了变形菌门中埃希菌、志贺菌等肠道致病菌的相对丰度。结论霍山石斛降低了IBS-D小鼠的内脏高敏感,可能通过恢复IBS-D小鼠肠道菌群的多样性,调节肠道菌群相对丰度,发挥对IBS-D的治疗作用。Objective To observe the effects of Dendrobium huoshanense on the gut microbiota of mice with diarrhea predominant-irritable bowel syndrome(IBS-D),and provide a basis for the study of the therapeutic mechanism of Dendrobium huoshanense on IBS-D.Methods Twenty-four SPF grade male mice were randomly divided into normal control group,model group,Dendrobium huoshanense group,and pinaverium bromide group,with 6 mice in each group.The IBS-D model was prepared by gavage of senna leaves combined with restrained tail clamping stress.After successful modeling,the Dendrobium huoshanense group and pinaverium bromide group were given corresponding drug doses by gavage for one week.The general status,fecal characteristics,fecal moisture content,abdominal withdrawal reflex score were compared among groups;changes in gut microbiota species and abundance of mice in each group were detected using 16S rRNA gene high-throughput sequencing.Results Compared with the normal controls,the IBS-D model mice showed poor mental health,increased bowel movements,increased fecal moisture content and visceral sensitivity,and decreased diversity and abundance of gut microbiota.After intervention with Dendrobium huoshanense,compared with the model controls,the mental status of mice in the Dendrobium huoshanense group was improved,and the fecal characteristics returned to normal,while the fecal moisture content and visceral sensitivity decreased.The relative abundance and composition of species indicated that Dendrobium huoshanense increased the diversity of gut microbiota in IBS-D mice and significantly increased the relative abundances of beneficial bacteria genera such as norank_Muribaculaceae,Lactobacillus,Alloprevotella,and decreased those of pathogenic bacteria such as Escherichia and Shigella in the Proteobacteria phylum.Conclusion Dendrobium huoshanense reduce visceral hypersensitivity in IBS-D mice,possibly by restoring the diversity of gut microbiota in IBS-D mice,regulating the relative abundance of gut microbiota,and exerting therapeuti
