摘　　要：目的探讨传统中药黄连素对人膀胱癌5637细胞的抑制效应及可能机制。方法使用不同浓度的黄连素(20、40、80、160μmol/L)分别处理人膀胱癌5637细胞24、48、72 h,采用CCK-8实验检测黄连素对膀胱癌5637细胞的抑制效应并使用流式细胞术检测黄连素对癌细胞增殖周期的改变;通过qPCR、Western blotting实验检测黄连素对膀胱癌细胞中EZH2、CDK1 mRNA及蛋白表达水平的影响。结果经过黄连素处理后,膀胱癌5637细胞的增殖受到了明显抑制,抑制强度与黄连素处理的时间以及其药物浓度呈正相关;且黄连素可使肿瘤细胞G 0/G 1期受到明显阻滞,还能显著降低EZH2、CDK1 mRNA及蛋白的表达。结论黄连素可抑制膀胱癌细胞的生长并阻滞细胞周期,机制可能与药物黄连素下调膀胱肿瘤细胞中EZH2、CDK1的表达有关。Objective To study the effects of Berberine on the growth of human bladder cancer cell lines 5637,and to elucidate the possible mechanism of such effects.Methods The inhibitory effect of Berberine on cell proliferation was examined by CCK-8 and cell cycle was detected by flow cytometry(FCM).The expression levels of CDK1,EZH2 mRNA and protein levels were detected by qPCR and Western blotting.Results The Berberine significantly inhibited cell proliferation,and blocked the cell cycle in the G 0/G 1 phase.The expression levels of CDK1,EZH2 mRNA and protein down-regulated when 5637 cell were treated by Berberine.Conclusions Berberine significantly inhibited cell proliferation,and blocked the cell cycle in the G 0/G 1 phase.These effects may be related with its function in down-regulating the expression levels of CDK1 and EZH2 protein.
