基于系统中药学的附子回阳救逆功效标志物的挖掘

摘　　要：[目的]以系统中药学理论为指导,挖掘附子回阳救逆的功效标志物,并阐释其回阳救逆的机制。[方法]搜集附子主要化学成分和靶点,建立蛋白互作网络和生物模块分析方法,以“中药-成分-靶点-药理-功效”的模式,从元素、结构、边界、功能层面明确附子回阳救逆功效与其他功效的边界,阐释附子回阳救逆的机制,挖掘附子回阳救逆的功效标志物。[结果]次乌头原碱、多根乌头碱、查斯曼宁碱、去甲乌药碱、去氢宋果灵、附子吡嗪B、一枝蒿丙素、苯甲酰次乌头原碱、去甲猪毛菜碱、宋果灵、和厚朴酚、异甘草素这12个成分能通过9个关键靶点作用于6个功能模块,通过参与G蛋白偶联受体蛋白信号通路、程序性细胞死亡的调控、细胞内钾离子转运、类固醇合成、细胞内蛋白激酶级联、氧化还原反应发挥附子回阳救逆的功效。[结论]挖掘了附子回阳救逆的功效标志物,现有研究可以佐证筛选出的功效成分,可为附子的质量控制与药物研发提供新的思路与方法。[Objective]This study aims to explore efficacy markers and mechanisms of Radix Aconiti Lateralis Preparata in restoring yang and rescuing patient from collapse based on the theory of systematic traditional Chinese medicine.[Methods]We collected the main chemical components and targets of Radix Aconiti Lateralis Preparata,and established a protein interaction network and biological module analysis method.Following the pattern of“herb-component-target-pharmacology-efficacy”we clarified the boundaries of Radix Aconiti Lateralis Preparata’s efficacy in restoring yang and rescuing patient from collapse from the perspectives of elements,structures,boundaries,and functional levels.We also elucidated the mechanisms of Radix Aconiti Lateralis Preparata in restoring yang and rescuing patient from collapse,as well as identified efficacy markers.[Results]Twelve components,namely Aconitine,Benzoylmesaconine,Chasmanine,Deacetylaconitine,Delphinine,Fuzipyridine B,Artemisinin,Benzoylaconine,Deacetylpubescenine,Delphinine,Magnolol,and Isoliquiritigenin,targeted nine key proteins and acted on six functional modules.They exerted the efficacy of restoring yang and rescuing patient from collapse by participating in G protein-coupled receptor signaling pathways,regulating programmed cell death,intracellular potassium ion transport,steroid synthesis,intracellular protein kinase cascades,and redox reactions.[Conclusion]This study identified efficacy markers for Aconiti Lateralis Preparata in restoring yang and rescuing patient from collapse.The selected effective components,supported by existing research,can provide new ideas and methods for quality control and drug development of Aconiti Lateralis Preparata.
