摘 要:研究旨在探究紫花苜蓿水提物与山楂叶水提物复配对HepG2细胞脂肪堆积的影响及其抗氧化活性。以油酸体外诱导HepG2细胞建立脂肪堆积模型,并分别以紫花苜蓿提取物、山楂叶提取物、紫花苜蓿提取物-山楂叶提取物复配进行干预,采用MTT法测定细胞活力;油红O染色法观察脂滴分布;生物试剂盒测定细胞内三酰甘油(TG)、总胆固醇(TC)、丙二醛(MDA)含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及总抗氧化能力(T-AOC)。结果显示,紫花苜蓿提取物-山楂叶提取物复配(6∶4)复配可减少油酸诱导HepG2细胞所导致的脂滴数量增加,降低细胞中TG、TC含量。并增强抗氧化能力,可提高细胞内SOD、GSH-Px酶活性和总抗氧化能力(P<0.01),减少MDA的产生(P<0.05)。综上可得出结论,紫花苜蓿提取物-山楂叶提取物复配可显著抑制油酸诱导HepG2脂肪堆积,其作用优于紫花苜蓿提取物或山楂叶提取物单体,并可提高细胞内源性抗氧化能力。The experiment aim of this study was to investigate the effects of alfalfa water extract and haw⁃thorn leaf water extract on fat accumulation in HepG2 cells and their antioxidant activities.The adipose accumulation model of HepG2 cells was induced by sodium oleate in vitro.Alfalfa extract、hawthorn leaf extract、alfalfa extract-Hawthorn leaf extract combination were used to intervene,and cell viability was determined by MTT method.The distribution of lipid droplets was observed by oil red O staining.The contents of triglyceride(TG),total cholesterol(TC),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione oxidase(GSH-Px)and total an⁃tioxidant capacity(T-AOC)were determined by bioassay.The results showed that the combination of alfalfa extract and hawthorn leaf extract(6∶4)could reduce the number of lipid droplets induced by oleic acid in HepG2 cells,and reduce the contents of TG and TC in the cells.The combination of LC(6∶4)can significantly reduce the increase in the number of lipid droplets and the contents of TG and TC caused by sodium oleate induction in HepG2 cells,and has antioxidant capacity,and can increase the activity of SOD and GSH-Px enzymes and total antioxidant capacity in cells(P<0.01),decreased the production of MDA(P<0.05).In conclusion,the combination of alfalfa extract and hawthorn leaf extract can significantly inhibit the accumulation of HepG2 fat induced by sodium oleate,which is superior to the monomers of alfalfa extract or hawthorn leaf extract,at the same time it can improve the endogenous antioxidant capacity of cells.
