TiO_(2)负载血浆凝血酶的制备及其对血液快速促凝性能的研究

摘　　要：目的探究TiO_(2)负载血浆凝血酶的制备及其对血液快速促凝的作用,以及用于止血材料的可行性。方法通过在复钙血浆中加入TiO_(2)纳米颗粒,制备TiO_(2)/血浆凝血酶促凝血材料,并通过BET、FTIR、XRD等方式分析其组分结构,通过凝血试验证明这种材料对血液的促凝效果,并在常温下储存28d后,测试促凝效果变化。结果TiO_(2)/血浆凝血酶促凝材料具有良好的止血效果。凝血酶稳定地负载在TiO_(2)中,在室温下保存28d后,仍可保持较高的凝血活性。在细胞毒性试验中,浓度25~100μg/mL的TiO_(2)/血浆凝血酶均无明显细胞毒性。结论血浆凝血酶在TiO_(2)表面负载后,具有快速凝血性、稳定性和安全性。TiO_(2)/血浆凝血酶有望在机体快速止血方面发挥潜在作用。Objective To investigate the preparation and rapid coagulation blood performance of TiO_(2) loaded plasma thrombin,as well as the feasibility of using as hemostatic materials.Methods TiO_(2)/plasma thrombin promoting coagulation material was prepared by adding TiO_(2) nanoparticles into calcium compound plasma,and its component structure was further demonstrated by means of BET,FTIR,XRD,etc.The effect of this material on blood coagulation was proved by coagulation test,and changes in the procoagulant effect were tested after 28 d of storage at room temperature.Results TiO_(2)/plasma thrombin procoagulant had good hemostatic effect.The thrombin was stably loaded in TiO_(2) and maintained high coagulation activity after 28 d of storage at room temperature.In the cytotoxicity test,TiO_(2)/plasma thrombin showed no obvious cytotoxicity at 25~100μg/mL concentration.Conclusion Plasma thrombin has rapid coagulability,stability and safety after loading on TiO_(2) surface.TiO_(2)/plasma thrombin is expected to play a potential role in rapid hemostasis.
