荆防颗粒增强免疫作用机制研究

摘　　要：目的探讨荆防颗粒增强免疫作用及作用机制。方法采用模式生物斑马鱼,分别建立雷帕霉素致中性粒细胞减少、酒石酸长春瑞滨致巨噬细胞和胸腺T细胞减少的斑马鱼免疫低下模型,以中性粒细胞数目、巨噬细胞荧光强度、T细胞荧光强度以及γ干扰素(interferon-γ,IFN-γ)含量为药效评价指标,探究荆防颗粒增强免疫的药效作用。采用环磷酰胺免疫低下小鼠模型,以脏器指数、血清细胞因子、血清免疫球蛋白等指标评价荆防颗粒的免疫调节活性,采用qRT-PCR探究荆防颗粒的免疫调节作用机制。结果荆防颗粒可以增加中性粒细胞、巨噬细胞和T细胞数量及IFN-γ含量(P<0.05、0.01、0.001),增加环磷酰胺免疫低下小鼠的脏器指数(P<0.05),增加小鼠血清中白细胞介素-2(interleukin-2,IL-2)、肿瘤坏死因子-α(tumor necrosis factor,TNF-α)、免疫球蛋白G(immunoglobulin G,IgG)、免疫球蛋白M(immunoglobulin M,IgM)的含量(P<0.05、0.01、0.001),显著下调小鼠脾组织中核因子-κB(nuclear factor-κB,NF-κB)通路相关基因表达(P<0.05、0.01),从而起到增强免疫的作用。结论荆防颗粒具有增强免疫的作用,其作用机制与抑制NF-κB通路有关。Objective To explore the effect and mechanism of Jingfang Granules(荆防颗粒)on enhancing immunity.Methods A model organism zebrafish was used to establish immunocompromised models of rapamycin-induced neutropenia,vinorelbine tartrate-induced macrophage and thymic T cell reduction in zebrafish.The number of neutrophils,macrophage fluorescence intensity,T cell fluorescence intensity and interferon-γ(IFN-γ)content were used as a pharmacological evaluation index to explore the pharmacological effect of Jingfang Granules on enhancing immunity.Using a cyclophosphamide immunocompromised mouse model,the immune regulatory activity of Jingfang Granules was evaluated using indicators such as organ index,serum cytokines and serum immunoglobulin,qRT PCR was used to explore the immune regulatory mechanism of Jingfang Granules.Results Jingfang Granules could increase the numbers of neutrophils,macrophages and T cells,as well as IFN-γcontent(P<0.05,0.01,0.001),increase the organ index of immunocompromised mice with cyclophosphamide(P<0.05),increase levels of interleukin-2(IL-2),tumor necrosis factor-α(TNF-α),immunoglobulin G(IgG)and immunoglobulin M(IgM)in serum(P<0.05,0.01,0.001),and significantly down-regulate nuclear factor-κB(NF-κB)pathway related gene expressions in spleen tissue of mice(P<0.05,0.01),thereby playing a role in enhancing immunity.Conclusion Jingfang Granules has the effect of enhancing immunity,and its mechanism is related to the inhibition of NF-κB pathway.
