摘　　要：建立了高效液相色谱法测定大鼠血浆中奥拉帕利的血浆蛋白结合率,对血浆中奥拉帕利的含量进行测定,用平衡透析法进行奥拉帕利的血浆蛋白结合率测定。采用Kromasil Column C18色谱柱,流动相为甲醇∶水(50∶50),流速1.0 mL/min,检测波长为275 nm,应用平衡透析结合HPLC-UV法,对3个浓度的奥拉帕利大鼠的血浆蛋白结合率进行测定。在低、中、高浓度下与大鼠的血浆蛋白结合率分别为80.09%、78.44%、79.06%,平均值为79.20%。研究结果表明,采用高效液相色谱法结合平衡透析法可以准确并快速地测得大鼠血浆中奥拉帕利的血浆蛋白结合率。The plasma protein binding rate of olaparib in rat plasma was determined by the high-performance liquid chromatography method,and the plasma protein binding rate of olaparib was determined by equilibrium dialysis.The plasma protein binding ratio of three concentrations of olaparib was measured using Kromasil Column C18 column with a mobile phase of methanol:water(50∶50),the detection wavelength of 275 nm,and equilibrium dialysis combined with HPLC-UV method.At low,medium and high concentrations,plasma protein binding to rats was 80.09%,78.44%,and 79.06%,respectively,with a mean value of 79.20%.The results show that the plasma protein binding rate of olaparib can be measured accurately and quickly measured by HPLC and equilibrium dialysis.
