芦荟胶屏障修护功效研究

摘　　要：初步探究芦荟胶的皮肤屏障修护功效和作用机制。通过体外细胞实验,研究芦荟胶对人永生化角质细胞和真皮成纤维细胞的细胞增殖和细胞迁移率的影响,并采用UVA紫外辐照损伤细胞模型,对活性氧自由基含量、皮肤屏障相关基因表达水平以及I型胶原蛋白分泌进行检测;采用人体临床测试,基于仪器测量和受试者自评等方面对其进行功效评估。结果显示,添加1 mg/mL~4 mg/mL芦荟胶能促进角质细胞和成纤维细胞的细胞增殖和迁移(P<0.01),能显著提高经紫外损伤的细胞存活率,同时降低了细胞内ROS含量(P<0.01);较高浓度的芦荟胶能显著提高FLG和Cldn-1表达水平,促进I型胶原蛋白分泌(P<0.01);使用芦荟胶28天后,受试者皮肤泛红、脱屑、干燥度、敏感度、灼热感及紧绷感较使用前皆有改善;受试者在使用产品4~6天皮肤角质层含水量显著上升,皮肤经表皮失水率、皮肤血红素以及泛红区面积显著下降(P<0.05)。综上,芦荟胶具有屏障修护功效,可能通过促进表皮细胞增殖和细胞愈合,抑制ROS的产生,促进屏障基因表达和胶原蛋白分泌,从而减少表皮细胞的损伤和凋亡共同实现。This study is to dissect the effect and mechanism of Aloe vera gel on barrier repair.The effects of Aloe vera on the cell proliferation and cell migration rate of human immortalized keratinocytes and dermal fibroblasts were studied by in vitro cell experiments.The cell model of UVA radiation injury was used to detect the ROS content,skin barrier related gene expression level and type I collagen secretion.Human clinical test was used for evaluation based on two dimensions:instrumental measurement and subject self-evaluation.The results showed that adding 1 mg/mL~4 mg/mL Aloe vera gel promoted the cell proliferation and migration of keratinocytes and dermal fibroblasts(P<0.01),significantly improved the survival rate of cells damaged by ultraviolet rays,and at the same time reduced the intracellular ROS content(P<0.01).Higher concentrations of Aloe vera gel significantly increased the expression levels of FLG and Cldn-1 genes,and promoted the secretion of type I collagen(P<0.01).After using Aloe vera gel for 28 days,the subjects skin redness,scaling,dryness,sensitivity,burning sensation and tightness were all improved compared to before use.After using the product for 4 to 6 days,the water content of the stratum corneum increased significantly,and TEWL value,skin hemoglobin,and red area significantly decreased(P<0.05).In conclusion,Aloe vera gel has barrier repair effects.This effect may be achieved by promoting epidermal cell proliferation and cell healing,inhibiting the production of ROS,promoting barrier gene expression and collagen secretion,thereby reduci ng epidermal cell damage and apoptosis.
