摘 要:[目的]探讨北沙参提取物对放射性肺损伤大鼠肺部炎症的影响。[方法]将大鼠随机分为6组:假手术组、模型组、阳性对照组及北沙参提取物低、中、高剂量组(18.75 mg/kg、37.50 mg/kg、75.00 mg/kg),每组12只,采用直线加速器单次照射15 Gy构建放射性肺损伤动物模型,照射完毕后连续给药28 d,造模结束后处死。肺组织进行HE、Masson染色评价其病理改变,ELISA法检测大鼠血清炎症因子,实时荧光定量PCR检测大鼠肺组织中炎症因子与炎症浸润调控关键蛋白的mRNA表达水平,蛋白免疫印迹法检测大鼠肺组织信号通路相关蛋白表达水平。[结果]HE染色显示,与假手术组比较,模型组肺泡炎症显著提高(P<0.05),各剂量组炎性细胞浸润减少(P<0.05);与假手术组比较,其余各组大鼠血清TGF-β1、TNF-α、IL-1β、IL-6水平,肺组织中TGF-β1、p-Smad2/3蛋白与IL-1β、IL-6、TNF-α、TGF-β1、α-SMA和COL1A1 mRNA表达水平均显著升高(P<0.05),而各干预组大鼠的表达水平较模型组均显著降低(P<0.05),且呈剂量反应关系(P<0.05)。[结论]北沙参提取物对急性放射性肺损伤具有保护作用,其作用机制可能与降低炎性因子、改变炎症调控蛋白表达密切相关。[Objective]To explore the effect of extract of Radix Adenophora on lung inflammation in rats with radiation lung injury.[Method]The rats were randomly divided into 6 groups:Sham operation group,model group,positive control group and low-dose,medium-dose and high-dose groups(18.75 mg/kg,37.50 mg/kg,75.00 mg/kg)of extract of Parapsychia palmata,with 12 animals in each group,were irradiated by linear accelerator at a single time of 15 Gy to establish radiation-induced lung injury animal models,and were given continuous administration for 28 days after irradiation,and killed after modeling.The pathological changes of the lung tissues were evaluated by HE and Masson staining,the serum inflammatory factors were detected by ELISA,the mRNA expression levels of inflammatory factors and key proteins in the regulation of inflammatory infiltration in the lung tissues were detected by real-time fluorescent quantitative PCR,and the expression levels of proteins related to signaling pathways in the lung tissues were detected by Western Blot.[Result]HE staining showed that compared with sham operation group,alveolar inflammation was significantly increased in model group(P<0.05),and inflammatory cell infiltration was decreased in each dose group(P<0.05).Compared with the sham operation group,the levels of TGF-β1,TNF-α,IL-1βand IL-6 in serum of rats in the other groups were as follows:The expression levels of TGF-β1,p-SMad2/3 protein and IL-1β,IL-6,TNF-α,TGF-β1,α-SMA and COL1A1 mRNA in lung tissues were significantly increased(P<0.05),while the expression levels of intervention groups were significantly decreased compared with model group(P<0.05).There was a dose-response relationship(P<0.05).[Conclusion]The extract of Radix Adenophora has protective effect on acute lung injury caused by radiation,and its mechanism may be closely related to reducing inflammatory factors and changing the expression of inflammatory regulatory proteins.
