摘 要:为了筛选与繁殖性能相关的显著差异表达蛋白及信号通路,试验采用TMT(tandem mass tag)标记蛋白定量技术与生物信息学分析方法,对高产(产仔数在11只以上)与低产(产仔数在7只以下)乌苏里貉卵巢组织进行差异表达蛋白筛选,GO功能注释、KEGG信号通路富集分析。结果表明:在乌苏里貉卵巢组织中共鉴定到4915种蛋白,其中372种为显著差异表达蛋白(P≤0.05,差异倍数≥1.2或≤0.83),上调307种,下调65种。差异表达蛋白主要参与了羧酸代谢过程、胞内氨基酸代谢过程、转录起始、胞内脂代谢过程、磷脂代谢过程、GTP生物合成过程、UTP生物合成过程、CTP生物合成过程等生物学过程,主要参与了组氨酸代谢、氨酰-tRNA生物合成、N-聚糖生物合成、β-丙氨酸新陈代谢、PI3K-Akt信号通路、mTOR信号通路等16条信号通路;有11种差异表达蛋白与卵泡发育和排卵相关。蛋白网络互作分析获得10种核心蛋白,参与了蛋白折叠和蛋白合成。说明试验筛选到的显著差异表达蛋白可能与乌苏里貉的产仔数相关。In order to screen for significantly differentially expressed proteins and signaling pathways related to reproductive performance,in the experiment,using TMT(tandem mass tag)quantification technology and bioinformatics analysis methods,the ovary tissues of high-breeding(more than 11 pups)and low-breeding(less than 7 pups)Ussuri raccoon dogs were screened for differentially expressed proteins,and GO functional annotation and KEGG signaling pathway enrichment were performed.The results showed that a total of 4915 proteins were identified in the ovary tissues of Ussuri raccoon dogs,of which 372 were significantly differentially expressed proteins(P≤0.05,fold difference≥1.2 or≤0.83),307 were up-regulated proteins and 65 were down-regulated proteins.The differentially expressed proteins were mainly involved in biological processes such as carboxylic acid metabolic process,intracellular amino acid metabolism,transcription initiation,intracellular lipid metabolism,phospholipid metabolism,GTP biosynthesis,UTP biosynthesis,and CTP biosynthesis,and they were mainly involved in 16 signaling pathways including histidine metabolism,aminoacyl-tRNA biosynthesis,N-glycan biosynthesis,β-alanine metabolism,PI3K-Akt signaling pathway,and mTOR signaling pathway.Eleven differentially expressed proteins were associated with follicular development and ovulation.Protein network interaction analysis obtained 10 core proteins,which were involved in protein folding and protein synthesis.The results suggested that these differentially expressed proteins might be related to the litter number of Ussuri raccoon dogs.
