通关藤总皂苷对LPS诱导的HepG2细胞炎症改善作用研究

摘  要:目的探讨通关藤总皂苷(MaT)对脂多糖(LPS)诱导的HepG2细胞炎症模型的抑制作用。方法采用四氮唑盐(MTT)法、酶联免疫吸附法(ELISA)筛选出最适LPS造模浓度和诱导时间,建立LPS诱导的HepG2细胞炎症模型;MTT法、ELISA法筛选出MaT干预炎症HepG2细胞浓度范围。采用反转录实时定量聚合酶链式反应RT-PCR技术,检测不同浓度MaT干预后炎症信号通路STAT3和NF-κB通路相关基因的表达。结果以2μg/mL LPS与HepG2细胞在37℃,5%CO_(2)的条件下共培养24 h,建立LPS诱导的HepG2细胞炎症模型。相较于正常组,模型组白介素-6(IL-6)、白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)的分泌水平显著提高(P<0.01);MaT干预后,IL-6、IL-8、TNF-α浓度呈剂量依赖性下降,并且下调了炎症信号通路中STAT3、MCL-1、XIAP、BCL-2、NF-κB mRNA的表达。结论通关藤总皂苷对LPS诱导的HepG2细胞炎症模型具有抑制作用,其机制可能为通关藤总皂苷通过抑制STAT3/NF-κB信号通路相关基因表达,减少炎症因子的释放有关。Objective To investigate the inhibitory effect of total saponins of Tongguanteng(Marsdeniae Tenacissimae Caulis)(MaT)on lipopolysaccharide(LPS)-induced inflammation in HepG2 cells.Methods The optimal LPS modeling concentration and induction time were screened by tetrazolium salt(MTT)method and ELISA method,and the LPS-induced HepG2 cell inflammation model was established.RT-PCR technology was used to detect the expression of STAT3 and NF-κB pathway-related genes in the inflammatory signaling pathway after intervention with different concentrations of MaT.Results LPS and HepG2 cells were co-cultured with 2μg/mL LPS for 24 h at 37℃and 5%CO_(2) to establish an LPS-induced HepG2 cell inflammation model.Compared with the normal group,the secretion levels of interleukin-6(IL-6),interleukin-8(IL-8),and tumor necrosis factor-α(TNF-α)were significantly increased in the model group(P<0.01);After MaT intervention,the concentrations of IL-6,IL-8,and TNF-αdecreased in a dose-dependent manner,and the expression of STAT3,MCL-1,XIAP,BCL-2,and NF-κ-B mRNA in the inflammatory signaling pathway was downregulated.Conclusion The total saponins of Tongguanteng(Marsdeniae Tenacissimae Caulis)have an inhibitory effect on the LPS-induced HepG2 cell inflammation model,and the mechanism may be related to the reduction of the release of inflammatory factors by inhibiting the expression of genes related to the STAT3/NF-κB signaling pathway.
