摘 要:利用合成的芥子气-氨基酸加合物及其同位素标记化合物,基于超高效液相色谱-同位素稀释串联质谱技术,建立了芥子气染毒人血浆中5种氨基酸加合物的绝对定量分析方法。5种氨基酸加合物在0.100~100 ng/mL范围内线性良好,相关系数R2>0.995,检出限分别为0.0200 ng/mL([N1-HETE]-His和[N3-HETE]-His)和0.0500 ng/mL(HETE-Lys,HETE-Asp和HETE-Glu)。应用该定量方法研究了芥子气在人血浆蛋白上不同加合位点的反应趋势。本文所建立的氨基酸加合物绝对定量方法,不仅提供了一种芥子气中毒快速溯源方法,还为芥子气暴露人员的临床诊断救治及代谢机理研究等提供了重要的手段。An absolute quantitation method for the determination of 5 amino acid adducts in sulfur mustard-exposed plasma through ultra-high performance liquid chromatography-isotope-dilution tandem mass spectrometry was developed with the synthesized reference adducts.This method for the detection of 5 amino acid adducts showed good linearity in the concentration range of 0.100-100 ng/mL,along with the correlation coefficient(R~2)higher than 0.995.The limits of detection were 0.0200 ng/mL for[N1-HETE]-His and[N3-HETE]-His and 0.0500 ng/mL for HETE-Lys,HETE-Aspand HETE-Glu.This quantitative method has been applied in the study of reactivity between adduction sites on human plasma protein and sulfur mustard.The absolute quantitation method of amino acid adducts can provide not only a rapid approach for retrospective determination of sulfur mustard exposure,but also an important tool for clinical diagnosis and treatment of sulfur mustard-exposed individuals and metabolic mechanism study.
