摘 要:猪传染性胃肠炎病毒(transmissible gastroenteritis virus,TGEV)主要感染小肠上皮细胞,引起不同年龄的猪呕吐、腹泻和脱水,对2周龄以内的仔猪致死率高,其致病性与肠黏膜屏障的完整性密切相关。而紧密连接蛋白对肠黏膜屏障的维持和修复至关重要,本试验旨在探究TGEV对紧密连接蛋白Claudin-4和Claudin-5表达的影响。通过检测Claudin-4和Claudin-5在SPF猪小肠组织和大肠组织的分布情况,以及采用RT-qPCR法检测TGEV感染后仔猪肠道组织及体外细胞中Claudin-4和Claudin-5mRNA含量的变化,结果显示:Claudin-4和Claudin-5在十二指肠中含量最高,在盲肠中含量最低,且TGEV感染后仔猪肠道组织及体外细胞中Claudin-4和Claudin-5mRNA水平均显著上调。综上所述,研究发现了紧密连接蛋白Claudin-4和Claudin-5在猪肠道组织内的分布情况,验证了TGEV可以上调紧密连接蛋白Claudin-4和Claudin-5的表达,为深入探讨Claudin-4和Claudin-5与TGEV间的作用机制奠定了基础,也为制定TGEV新的防控策略提供了思路。The transmissible gastroenteritis virus(TGEV)mainly infects small intestinal epithelial cells,causing vomiting,diarrhea,and dehydration in pigs of different ages.It has a high mortality rate for piglets under two weeks of age,and its pathogenicity is closely related to the integrity of the intestinal mucosal barrier.Tight junction proteins are crucial for maintaining and repairing the intestinal mucosal barrier.This article aims to investigate the effect of TGEV on the expression of tight junction proteins Claudin-4and Claudin-5.The distribution of Claudin-4and Claudin-5in the small intestine and large intestine of SPF pigs,as well as the changes in mRNA expression of Claudin-4and Claudin-5in the intestinal tissue and in vitro cells of piglets infected with TGEV were detected using RT-qPCR,the results showed that Claudin-4and Claudin-5had the highest abundance in the duodenum and the lowest abundance in the cecum.After TGEV infection,the levels of Claudin-4and Claudin-5mRNA in the intestinal tissue and in vitro cells of piglets were significantly upregulated.In summary,this study found the distribution of tight junction proteins Claudin-4and Claudin-5in pig intestinal tissue and verified that TGEV can upregulate the expression of tight junction proteins Claudin-4and Claudin-5.This study laid the foundation for in-depth exploration of the mechanism of action between Claudin-4,Claudin-5and TGEV,and provided ideas for developing new prevention and control strategies for TGEV.
