摘 要:骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)是具有自我更新及多向分化潜能的成体干细胞,在维持骨组织稳态、促进骨损伤修复中发挥重要作用。衰老是机体随着生命周期延长发生的结构和功能上的不可逆变化,干细胞衰老是其重要原因之一。BMSCs的衰老不仅会导致骨稳态失衡以及骨质疏松等疾病,还会使BMSCs的临床应用受到限制。线粒体是细胞内进行能量代谢的重要细胞器,近年来研究表明线粒体功能障碍在衰老的发生和发展过程中发挥重要作用。本文对线粒体调控BMSCs衰老的作用机制进行系统综述,以期为防治BMSCs衰老、诊治衰老和代谢性骨疾病提供新的思路。Bone marrow mesenchymal stem cells(BMSCs)possess self-renew and multipotent differentiation capability and play important roles in maintaining bone homeostasis and injury recovery.Aging is an irreversible process of structural and functional changes that occur over lifespan in organisms,and stem cells senescence is an essential mechanism underlying aging.BMSCs senescence may result in skeletal disorders such as osteoporosis and significantly restrict the application of BMSCs transplantation in clinic.Mitochondria control energy metabolism in cells and mitochondria dysfunction has been found to play an important role in aging and aging-related diseases.In this review,we summarize the recent advances of the relationship between mitochondria dysfunction and BMSCs senescence.We hope the review may provide prospective strategies for the therapies of BMSCs senescence and aging-related metabolic bone disorders.
