红斑丹毒丝菌对猪脐静脉内皮细胞的感染及细胞增殖凋亡的影响

摘　　要：猪感染红斑丹毒丝菌后会表现出明显的皮肤和血管病变,为了解红斑丹毒丝菌感染后对血管内皮细胞的影响,本研究建立了红斑丹毒丝菌体外感染猪脐静脉内皮细胞模型;利用革兰染色、菌落平板培养法等检测红斑丹毒丝菌对猪脐静脉内皮细胞的黏附能力;采用CCK-8、Ed U、Annexin V-PE等试剂盒分别检测红斑丹毒丝菌对血管内皮细胞的活力、增殖及凋亡的影响。结果显示,红斑丹毒丝菌易黏附和侵入猪脐静脉内皮细胞,损伤细胞活力,抑制其细胞增殖并显著诱导其凋亡。本文揭示了红斑丹毒丝菌感染可能会通过抑制猪血管内皮细胞增殖并诱导其凋亡而导致机体血管系统病变的机制,为猪丹毒的预防及治疗提供了参考。Porcine infected with Erysipelothrix rhusiopathiae exhibits remarkable skin and vascular lesions.In order to disclose the effects of Erysipelothrix rhusiopathiae infection on the viability of vascular endothelial cells,an in vitro model of Erysipelothrix rhusiopathiae infection on porcine umbilical vein endothelial cells was established in this study.The adhesion ability of Erysipelothrix rhusiopathiae to porcine umbilical vein endothelial cells was measured by gram staining and colony plate culture.The effects of Erysipelothrix rhusiopathiae on cell viability,proliferation,and apoptosis were assessed using CCK-8,Ed U,and Annexin V-PE kits.Results from this study showed that Erysipelothrix rhusiopathiae efficiently adhered to and invaded into porcine umbilical vein endothelial cells.The viability and proliferation of porcine umbilical vein endothelial cells were significantly decreased by Erysipelothrix rhusiopathiae infection,while the cell apoptosis ratio was significantly elevated.This study reveals that infection with Erysipelothrix rhusiopathiae may lead to vascular system damage in porcine by inhibiting the proliferation of porine endothelial cells and inducing cell apoptosis.The findings will provide some new insights into the prevention and treatment of Erysipelothrix rhusiopathiae infection.
