摘  要:本研究旨在探究低氧诱导因子2α(HIF-2α)、瘦素(LP)及其受体(LEPR)在不同年龄牦牛肺组织中的分布特点,以及在牦牛肺组织生长发育过程中可能发挥的作用。本试验采用免疫组织化学染色、qRT-PCR和Western-blot法,对HIF-2α、LP及LEPR在不同年龄牦牛肺中的分布及表达水平进行探究。免疫组化结果显示,HIF-2α主要表达在不同年龄牦牛肺终末细支气管上皮及伴行的肺动脉内皮;LP和LEPR表达部位相似,主要分布在不同年龄牦牛肺的各级支气管上皮、肺动脉内皮及肺泡上皮。qRT-PCR结果显示,HIF-2α基因在3日龄牦牛肺中的表达量显著高于6月龄、3岁和6岁的牦牛(P<0.05);LP基因在3日龄和6岁牦牛肺中的表达量最高,显著高于6月龄的牦牛(P<0.05);LEPR基因在3日龄牦牛肺中的表达量显著高于6月龄、3岁和6岁的牦牛(P<0.05)。Western-blot结果显示,LP蛋白在3日龄牦牛肺中表达量最高,且显著高于其他年龄段牦牛(P<0.05);LEPR与HIF-2α蛋白在3日龄牦牛肺的表达显著高于6月龄和3岁牦牛的表达量(P<0.05),与6岁牦牛之间的差异不显著(P>0.05)。结果表明,HIF-2α、LP及LEPR在牦牛肺发育过程中可能发挥重要作用,本研究为进一步探究低氧环境下牦牛肺的适应机制提供一定的基础资料。The aim of this study was to investigate the distribution characteristics of hypoxia-inducible factor 2α(HIF-2α),leptin(LP)and its receptor(LEPR)in the lungs of yaks of different ages,and the possible roles they may play in the growth and development of yak lungs.In this experiment,immunohistochemical staining,qRT-PCR and Western-blot were used to investigate the distribution and expression levels of HIF-2α,LP and LEPR in the lungs of yaks of different ages.Immunohistochemistry showed that HIF-2αwas mainly expressed in the terminal bronchial epithelial cells and the accompanying pulmonary artery endothelium of yak lungs at different ages;LP and LEPR had similar sites of expression,and were mainly distributed in the bronchial epithelium at all levels of the lungs of yak lungs at different ages,pulmonary artery endothelium,and alveolar epithelium.qRT-PCR showed that the expression level of HIF-2αwas significantly higher(P<0.05)than that of 6-year-old,3-year-old,and 6-year-old yaks in 3-year-old and 6-year-old yak lungs.The qRT-PCR results showed that the expression of HIF-2αgene in the lungs of 3-day-old yaks was significantly higher than that in the lungs of 6-month-old,3-year-old and 6-year-old yaks(P<0.05),the expression of LP gene in the lungs of 3-day-old and 6-year-old yaks was the highest and significantly higher than that in 6-month-old yaks(P<0.05),and the expression of LEPR gene in the lungs of 3-day-old yaks was significantly higher than that in the lungs of 6-month-old,3-year-old and 6-year-old yaks(P<0.05).The results of Western-blot showed that LP protein had the highest expression at 3 days of age and was significantly higher than other ages(P<0.05).LEPR and HIF-2αproteins were significantly higher at 3 days of age than at 6 months of age and 3 years of age(P<0.05),and the difference was not significant between them and 6 years of age(P>0.05).The results indicated that HIF-2α,LP and LEPR might play important roles in the development of yak lungs,and the present study provides some basic infor
