摘　　要：目的建立以梯度洗脱测定市售雷公藤多苷片中甲素含量的方法。方法采用HPLC以甲醇-水为流动相梯度洗脱;流速为0.9ml/min;柱温为25℃;检测波长为218 nm。结果线性范围为0.65~41.68μg/ml,r=0.9998,平均回收率103.43%,RSD=0.96%(n=9)。结论梯度洗脱法比现行质量标准的检测方法重复性更好、精密度更高。可作为雷公藤制剂中甲素的定量检测方法。Objective To establish a method to determine the content of triptolide in Tripterygium glycosides tablets by HPLC with gradient elution.Methods The high performance liquid chromatography external standard method was adopted with methanol aqueous solution with gradient elution at a flow rate of 0.9 ml/min,column temperature of 25℃and detection wavelength of 218 nm.Results The linear relationship was good(r=0.9998)in the range of 0.65 to 41.68μg/ml with the average recovery of 103.43%and the RSD of 0.96%(n=9).Conclusion Compared to the detection method of triptolide content in the drug standard,the gradient elution is better in reproducibility and higher in accuracy.It can be used as a quantitative method for the quantitative analysis of triptolide in Tripterygium glycosides tablets.
