摘 要:朱顶红褪绿环斑病毒(hippeastrum chlorotic ringspot orthotospovirus, HCRV)是正番茄斑萎病毒属(Orthotospovirus)病毒,危害农艺作物。NSm蛋白是HCRV非结构蛋白,在同属其他病毒中被证明是病毒运动蛋白并且是宿主抗性基因的无毒因子,但在HCRV中NSm的作用并不明确。本研究运用生物信息学分析法对HCRVNSm蛋白的理化性质、空间结构和蛋白质以及相关功能进行了分析和预测。结果表明,Orthotospovirus各病毒的NSm序列在演化过程中相似性和保守性不高;HCRV NSm蛋白属于3A超家族,为碱性的稳定亲水性蛋白,无信号肽和切割位点,无跨膜区;其蛋白质二级结构主要是不规则的卷曲,功能预测结果表明,NSm可与ATP结合,与同种蛋白聚合,并参与催化反应,并涉及脂肪酸β氧化、DNA复制等过程。本研究将为研究HCRV的致病机制提供重要参考,为NSm的作用机制和深入分析HCRV的危害及快速诊断其流行原因提供重要参考。Hippeastrum chlorotic ringspot orthotospovirus(HCRV)is a virus of the genus Orthotospovirus,which harm to agricultural crops.NSm protein is a non-structural protein of HCRV.It has been proved to be a viral movement protein and a Avr factor of host resistance genes in other viruses of the same genus.However,the role of NSm in HCRV is not clear.In this study,bioinformatics analysis methods were used to analyze and predict the physical and chemical properties,spatial structure and protein-related functions of the HCRV NSm protein.The results showed that the NSm sequences of the Orthotospovirus viruses were low similarity and conservation during the evolution process;the HCRV NSm protein belonged to the 3A superfamily,and was basic and stable hydrophilic protein without signal peptides,cleavage sites,and no span Membrane region;the protein secondary structure was mainly random coils.The function prediction results showed that NSm could combine with ATP,polymerize with the same protein,and participate in catalytic reactions,and involved in fatty acidβoxidation,DNA replication and other biological processes.The results of this study will provide important references for studying the pathogenic mechanism of HCRV.
