摘 要:以台州湾滩涂湿地不同围垦年限(13 a,36 a,57 a)围垦区种植西兰花农田土壤为材料,采用高通量测序技术,探索不同围垦年限滩涂湿地土壤细菌群落结构和多样性指数的变化,以期为滩涂围垦区农作物合理规划和优化其耕作栽培技术提供依据。结果表明,所有样本基因组经16S测序共获得有效序列读数为126355.333条,用于物种分类的OTUs 7660个,归属于23门、82纲、148目、276科、651属。西兰花根际土壤细菌菌群丰富度指数Chao1和群落多样性指数(Shannon)以围垦36年的土壤样品为最高,其次是围垦57年的土壤样品,而以围垦13年的土壤样品最低,且不同围垦年限间其值差异显著(P<0.05)。研究区土壤细菌的优势菌门为变形菌门、酸杆菌门、拟杆菌门、放线菌门、蓝细菌门。研究区土壤细菌的优势菌纲为α-变形菌纲、γ-变形菌纲、β-变形菌纲、放线菌纲、酸杆菌纲、芽单胞菌纲。随着围垦年限的延长,土壤细菌变形菌门、酸杆菌门、放线菌门的相对丰度呈先上升再降低的趋势,土壤蓝细菌门的相对丰度呈升高的趋势,而土壤芽单胞菌门的相对丰度在不断地减少。In order to reveal the changes of soil quality after reclamation and provide a theoretical basis for the scientific regulation of the soil ecosystem,the changes of bacterial community structure in rhizosphere soil of broccoli collected from coastal reclamation area with different years of reclamation(13 a,36 a,57 a)in Taizhou estuary were analyzed by using the Illumina Miseq high-throughput sequencing technology.The results showed that the total of 126355.333 effective sequence readings and 7660 OTUs were obtained from nine soil samples based on high-throughput sequencing of the 16S gene.Bacterial species detected in these samples covered 23 phylum,82 class,148 orders,276 families,651 gene.The abundance and diversity of bacterial communities in soil rhizosphere of broccoli collected from coastal reclamation areas with 36 years of reclamation was the highest among all soil samples,followed by soil samples of 57 years of reclamation,while that of 13 years of reclamation was the lowest among all soil samples.The dominant phyla were Proteobacteria,Acidobacteria,Bacteroidetes,Actinobacteria and Cyanobacteria in all soil samples.At the class level,Alphaproteobacteria,Gammaproteobacteria,Betaproteobacteria,Actinobacteria,Acidobacteria and Gemmatimonadetes were the predominant bacterial communities existed in all the soil samples.The long term land reclamation had significant effects on soil bacteria communities compositions.With increasing of reclamation,relative abundances of Proteobacteria,Acidobacteria,Actinobacteria initially increased and then decreased,while the relative abundance of Cyanobacteria was characterized by increase trend and that of Gemmatimonadetes was characterized by decrease trend.
