摘  要:利用CRISPR/Cas9技术对一个在花粉中特异表达且编码玉米果胶甲酯酶抑制剂基因GRMZM2G451028进行基因编辑,获得了编码区322~323位碱基缺失2 bp的移码突变体,命名为pms1。结果表明,与野生型相比,pms1纯合突变体在春秋两季花粉育性和散粉能力均极显著降低,且在秋季高温条件下的育性极显著低于春季;pms1杂合突变体的花粉育性与野生型相比没有显著差异,说明pms1纯合突变体对温度较为敏感。与野生型相比,pms1纯合突变体的雄穗分枝数、播种至开花及散粉的天数显著减少,株高、穗位高、穗长、穗粗没有差异。CRISPR/Cas9 technology was used to edit the gene GRMZM2G451028,which is specifically ex⁃pressed in pollen and encodes a pectin methylesterase inhibitor,and resulting in a frame shift mutant(named pms1)with a 2 bp deletion at 322-323 bp in the coding region of the gene.Compared with the wild-type plants,both the pollen shedding ability and pollen fertility in the homozygous pms1 plants were significantly reduced in spring and autumn.In addition,pollen fertility in the homozygous pms1 under high temperature condition of autumn was signifi⁃cantly lower than that in spring.Pollen fertility in the heterozygous pms1 showed no significant difference compared to wild-type plants,indicating that the homozygous pms1 is sensitive to temperature.There were no significant dif⁃ferences in plant height,ear height,ear length and diameter between homozygous pms1 plants and wild-type plants.However,the number of tassel branches and the days from sowing to flowering or silking showed significant differ⁃ences.
