复发性流产患者ILC2与Th17/Treg比值的相关性研究

摘　　要：目的探讨复发性流产患者中2型固有淋巴细胞(ILC2)的作用及其与Th17/Treg比值的相关性。方法选取2022年10月至2023年10月新疆医科大学第一附属医院妇科中心门诊复发性流产患者30例为复发性流产组,另选取同期正常妊娠者30例为正常妊娠组、孕龄期体检正常且未孕女性30例为备孕期妇女组。采用流式细胞术检测各组外周血中ILC2占CD45+细胞比例、Th17和Treg细胞分别占CD4+T淋巴细胞比例,Pearson法分析复发性流产患者ILC2与Th17/Treg比值的相关性。结果与备孕期妇女组(0.634%±0.064%)和正常妊娠组(0.570%±0.028%)比较,复发性流产组(1.457%±0.887%)外周血中ILC2的比例明显升高,差异有统计学意义(P<0.01),正常妊娠组与备孕期妇女组比较,差异无统计学意义(P>0.05)。与正常妊娠组和备孕期妇女组比较,复发性流产组中Th17细胞占CD4+T细胞的比例明显增高,Treg细胞比例明显降低,差异有统计学意义(P<0.05),正常妊娠组与备孕期妇女组比较,Th17细胞和Treg细胞比例差异均无统计学意义。相关性分析显示复发性流产组ILC2与Th17/Treg比值呈正相关(r=0.798,P<0.001)。结论复发性流产患者外周血中ILC2和Th17细胞比例升高,Treg细胞比例降低,且ILC2与Th17/Treg比值呈正相关,这提示ILC2参与复发性流产的发生发展,影响正常妊娠,且复发性流产可能与Th17/Treg失衡有关。Objective To explore the role of type 2 inherent lymphocytes(ILC2)in recurrent abortion and its correlation with the Th17/Treg ratio.Methods Selected 30 patients with recurrent abortion in the Outpatient Department of the Gynecology Center of the First Affiliated Hospital of Xinjiang Medical University from October 2022 to October 2023 as the recurrent abortion group,and another 30 normal pregnant women as the normal pregnancy group,30 women with normal gestational age and no pregnancy during the same period as the pregnancy⁃preparing group.Flow cytometry was used to detect the proportion of ILC2 in CD45+cells,Th17 and Treg cells accounted for CD4+T lymphocytes in the peripheral blood of each group,and the correlation of ILC2 and the ratio of Th17/Treg in patients with recurrent abortion by Pearson method.Results Compared with the pregnancy⁃preparing group(0.634%±0.064%)and the normal pregnancy group(0.570%±0.028%),the proportion of ILC2 in peripheral blood in the recurrent abortion group(1.457%±0.887%)was significantly increased,and the difference was statistically significant(P<0.01).Comparing the normal pregnancy group and the pregnancy⁃preparing group,the difference was not statistically significant(P>0.05).Compared with the normal pregnancy group and the pregnancy⁃preparing group,the proportion of Th17 cells in CD4+T cells in the recurrent abortion group had increased significantly,the proportion of Treg cells decreased significantly,and the difference was statistically significant(P<0.05),while the difference of the proportion of Th17 cells and Treg cells between the normal pregnancy group and the pregnancy⁃preparing group had no statistical significance.The correlation analysis showed that ILC2 was positively correlated with the Th17/Treg ratio in the recurrent abortion group(r=0.798,P<0.001).Conclusion The proportion of ILC2 cells and Th17 cells in the peripheral blood of patients with recurrent abortion increases,the proportion of Treg cells decreases,and the ratio of ILC2 to Th17/Treg is po
