摘 要:锌离子(Zn^(2+))作为一种对人体至关重要的微量金属元素,在维持人体正常生理功能、能量转换、新陈代谢和信号传递等方面扮演着关键角色,人体内Zn^(2+)的检测和监控对于预防和治疗相关疾病具有非常重要的意义。利用萘并[2,1-b]呋喃-2-碳酰肼与4-(二乙氨基)水杨醛进行缩合,设计合成了比色响应Zn^(2+)的荧光分子探针(SPA)。探针SPA识别Zn^(2+)后荧光大幅增强且波长发生红移。利用HR-MS考察了探针SPA对Zn^(2+)的响应机制。探针SPA识别Zn^(2+)的检测限可低至1.13 nmol/L。探针SPA已成功应用于HeLa细胞中Zn^(2+)的共聚焦成像以及活鼠体内Zn^(2+)的荧光成像。Zinc ion(Zn^(2+)),as a vital trace metal element for the human body,playing a key role in maintaining the normal physiological function of the human body,energy conversion,metabolism and signal transmission,etc.The detection and monitoring of Zn^(2+)in the body was of great significance for the prevention and treatment of related diseases.A colorimetric responsive fluorescent probe(SPA)for Zn^(2+)was designed and synthesized by condensation of naphthalene and[2,1-b]furan-2-carbohydrazide with 4-(diethylamino)salicyl aldehyde.The fluorescence of probe SPA was greatly enhanced and the wavelength was obviously red-shifted after the identification of Zn^(2+).The response mechanism of probe SPA to Zn^(2+)was investigated by HR-MS.The detection limit of Zn^(2+)recognized by probe SPA can be as low as 1.13 nmol/L.The probe SPA has been successfully applied to confocal imaging of Zn^(2+)in HeLa cells and fluorescence imaging of Zn^(2+)in live mice in vivo.
