摘 要:在过去5000多年的驯化过程中,家蚕茧丝的产量是人们努力改进的重要指标之一。研究发现,在家蚕BmN细胞中过表达非受体酪氨酸蛋白激酶基因BmAbl1时,部分谷胱甘肽S转移酶(glutathione S-transferase)编码基因的表达水平显著上升。基于piggyBac转座系统构建了过表达BmAbl1的转基因家蚕品系,发现其与对照品系家蚕相比,5龄幼虫的体重显著增加,全茧量、蛹重、茧层量以及茧层率均显著上升。且后部丝腺中谷胱甘肽S转移酶编码基因的转录水平显著上调,丝素重链和丝素轻链基因表达量分别是对照品系的3.3倍和1.3倍。上述结果表明,过表达BmAbl1提高了家蚕的茧丝产量。During the past 5000 years of domestication,the yield of cocoon silk was one of the most important characteristics that people want to improve.It was found that the expression levels of some glutathione S-transferase-encoding genes increased significantly when BmAbl 1 was overexpressed in BmN cells of silkworm.In this study,a transgenic silkworm strain overexpressing BmAbl 1 based on the piggyBac transposon system was constructed.The results showed that compared with the control strain of silkworms,the weight of the fifth instar larvae of the BmAbl 1 overexpression strain increased significantly.Meanwhile,the cocoon weight,pupa weight,cocoon shell weight and cocoon shell rate all increased significantly.The transcriptional levels of glutathione S-transferase-encoding genes in the posterior silk gland of BmAbl 1 overexpression strain were significantly upregulated,and the expression levels of fibroin-heavy chain and fibroin-light chain genes were 3.3 times and 1.3 times higher than those of the control strain,respectively.These results showed that the overexpression of BmAbl 1 promoted the cocoon silk yield of silkworm.
