摘  要:为了研发灵敏、准确的非洲猪瘟快速诊断试纸产品,本研究基于免疫层析技术原理,以非洲猪瘟病毒(ASFV)p72蛋白三聚体为检测抗原,量子点(QDs)为标记材料,金黄色葡萄球菌A蛋白(SPA)为检测线,p72单克隆抗体为质控线,建立了非洲猪瘟病毒抗体荧光免疫层析试纸,并对试纸的检测性能进行鉴定。结果:荧光免疫层析试纸检测ASFV标准阳性血清的效价为1∶102400,与猪瘟病毒、口蹄疫病毒、伪狂犬病病毒、猪繁殖与呼吸道综合征病毒及猪圆环病毒2型阳性血清抗体无交叉反应,在临床样本检测中与商业化ASFV抗体检测试剂盒的符合率为92.92%。综上,该试纸具有良好的灵敏度、特异性、准确性及稳定性,可用于临床样品中ASFV抗体的快速筛查。Based on the principle of immunochromatography,water-soluable quantum dots(QDs)were used as the labeling material conjugated with the trimer of ASFV protein,Staphylococcus aureus protein A and ASFV p72 monoclonal antibody immobilized on the strip were used as the detection line and quality control line,respectively,and a sensitive and accurate test strip for rapid detection of African swine fever was established in this study.The results showed that the sensitivity of the immunochromatographic test strip was 1∶102400,with no cross-reaction with antibody positive sera of swine fever virus,swine foot-and-mouth disease virus,pseudorabies virus,porcine reproductive and respiratory syndrome virus and porcine circovirus type 2.The total coincidence rate of the strip with the commercial ELISA kit was 92.92%.This strip had good sensitivity,specificity,accuracy and stability,which might be used for rapid screening of African swine fever virus specific antibodies.
