摘 要:脱细胞外基质(decellularized extracellular matrix,dECM)材料在组织工程和再生医学中具有重要应用,超临界CO_(2)流体(supercritical CO_(2)fluid,SCF-CO_(2))技术已被证明可用于dECM材料的制备。文中综述了SCF-CO_(2)的基本原理、SCF-CO_(2)脱细胞及其灭菌机制,介绍了辅助溶剂的使用与预处理技术对SCF-CO_(2)脱细胞的影响,以及dECM材料的生物医学应用。结果表明:与常规方法相比,具有高渗透性、高扩散性的SCF-CO_(2)能更有效地保持细胞外基质(extracellular matrix,ECM)的结构完整性和生物活性,同时展现出高效灭菌的优势,且制备的dECM材料在组织修复和再生医学领域具有显著应用潜力。Decellularized extracellular matrix(dECM)materials play a significant role in tissue engineering and regenerative medicine.Supercritical CO_(2)fluid(SCF-CO_(2))technology has been proven to be applicable in the preparation of dECM materials.By reviewing relevant literature in recent years,this paper summarizes the basic principles of SCF-CO_(2),the mechanisms of decellularization and sterilization by SCF-CO_(2),and discusses the impact of auxiliary solvents and pretreatment techniques on SCF-CO_(2)-assisted decellularization,as well as the biomedical applications of dECM materials.The results show that,compared with conventional methods,SCF-CO_(2)with high permeability and high diffusivity could maintain the structural integrity and biological activity of extracellular matrix(ECM)more effectively and show the advantage of high sterilization efficiency.Moreover,the prepared dECM materials have significant potential for application in the field of tissue repair and regenerative medicine.
