驻车丸对TNBS诱导克罗恩病大鼠治疗作用的实验研究

摘　　要：目的:观察驻车丸对克罗恩病模型大鼠的治疗作用。方法:取SPF级SD大鼠40只,随机分为空白对照组、模型对照组、阳性对照组和驻车丸治疗组,每组10只。除空白对照组外,其他3组采用TNBS/乙醇诱导法建立克罗恩病大鼠模型。造模成功后,阳性对照组和驻车丸治疗组分别灌胃给予柳氮磺吡啶(500 mg/kg)和驻车丸混悬液(2.835 g/kg),空白对照组和模型对照组灌胃等量生理盐水。连续给药7 d后通过评估大鼠疾病活动指数(DAI)、结肠黏膜损伤指数(CMDI)和结肠组织学损伤评分(HS)来观察驻车丸对克罗恩病大鼠的治疗作用。结果:模型对照组大鼠在造模后7 d内的DAI评分均高于空白对照组,且差异均有显著性,P<0.01;阳性对照组及驻车丸治疗组大鼠DAI评分随着给药时间的延长而呈现降低趋势,自给药第3天开始两组DAI评分均显著低于模型对照组,P<0.01。模型对照组大鼠CMDI、HS评分显著高于空白对照组(P<0.01);阳性对照组和驻车丸治疗组经治疗,大鼠结肠CMDI、HS评分均显著低于模型对照组(P<0.01)。结论:驻车丸对克罗恩病大鼠有较好的治疗效果。Objective To observe the therapeutic effect of Zhuche Pill on model rats with Crohn's disease(CD).Methods Total 40 SPF SD rats were randomly divided into blank control group,model control group,positive control group and Zhuche Pill treatment group,with 10 rats in each group.In addition to the blank control group,the model rats with CD were established by TNBS/ethanol induction in the other 3 groups.After the modeling was successful,the positive control group and the Zhuche Pill treatment group were given sulfasalazine(500 mg/kg)and Zhuche Pill suspension(2.835 g/kg)by intragastric administration,respectively,and the blank control group and the model control group were given the same amount of normal saline by intragastric administration.After continuous administration for 7 days,the therapeutic effect of Zhuche Pill on Crohn's disease was observed by evaluating disease activity index(DAI),colonmucosa damage index(CMDI)and histopathological score(HS).Results The DAI score of the model control group was higher than that of the blank control group within 7 days after modeling,and the difference was significant(P<0.01).The DAI scores of the positive control group and the Zhuche Pill treatment group showed a decreasing trend with the prolongation of administration time,and the DAI scores of the two groups were significantly lower than that of the model control group from the third day of administration(P<0.01).The CMDI and HS scores of the model control group were significantly higher than those of the blank control group(P<0.01).After treatment,CMDI and HS scores in the positive control group and Zhuche Pill treatment group were significantly lower than those in the model control group(P<0.01).Conclusion Zhuche Pill has a good therapeutic effect on rats with CD.
