摘 要:目的初步探究岭南道地药材广藿香中标志性成分广藿香醇(PA)在小肠缺血再灌注损伤中对肠道的保护作用,以便发掘广藿香新的药用价值。方法实验分为假手术组、模型组及PA低、中、高剂量组,PA各剂量组连续7 d预防性灌胃给予PA后,通过结扎肠系膜上动脉后再复通建立小肠缺血再灌注模型,假手术组行假手术,再灌注4 h后检测小肠中肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、脂多糖(LPS)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量,并按Chiu 6级评分法对小肠组织进行病理学半定量评分,同时通过TUNEL染色,计算细胞凋亡指数。结果与假手术组相比,模型组TNF-α、IL-6、LPS、SOD、MDA含量,组织病理学评分,细胞凋亡指数均明显升高(P<0.05或0.01),在PA保护性干预后,组织病变情况有所改善,低、高剂量组能明显降低小肠缺血再灌注损伤大鼠肠组织中的TNF-α、SOD和MDA含量,高剂量组还可降低IL-6含量及细胞凋亡指数,但PA对降低肠组织中LPS含量无明显作用。结论初步发现广藿香醇可抑制由缺血再灌注损伤引起的肠道细胞凋亡,并可降低机体的氧化应激压力及炎症因子水平,从而保护肠道组织。Objective To investigate the protective effect of patchouli alcohol,which is the principal ingredient of Lingnan genuine medicinal herb Pogostemon cablin(Blanco)Benth.on the intestine ischemia reperfusion(IIR)injury in rats,so as to explore the new medicinal value of Pogostemon cablin(Blanco)Benth.Methods Rats were randomly divided into sham-operated group,model group,low dose,middle dose and high dose groups of Patchouli alcohol(PA).After oral administration of PA in each PA dose groups for 7 days,intestine ischemia reperfusion model was established by ligating superior mesenteric artery and reperfusion.The sham-operated group underwent sham-operation.After 4 hours reperfusion,the concentration of TNF-α,IL-6,LPS,SOD,MDA in small intestine were detected,the histopathological examination was scored according to the Chiu 6 grading method and the apoptosis index was calculated by TUNEL staining.Results Compared with the sham-operated group,the concentration of TNF-α,IL-6,LPS,SOD and MDA in model group elevated significantly(P<0.05 or 0.01),additionally,histopathological score and apoptosis index also elevated obviously(P<0.01).After treated with PA,the tissue lesions of small intestine were improved.The low and high dose groups of PA could conspicuously reduce the concentration of TNF-α,SOD and MDA.Moreover,the high dose group could suppress the IL-6 secretion and the cell apoptosis.However,in terms of LPS reduction,PA didn′t have a significant effect.Conclusion It has been preliminarily found that PA can protect the intestine by inhibiting the cell apoptosis caused by IIR injury,reducing the oxidative stress and suppressing the production of inflammatory markers.
