畲药十二时辰醇提物外用对大鼠佐剂性关节炎的抗炎作用研究

摘　　要：目的:探讨畲药“十二时辰”醇提物外用对类风湿性关节炎模型大鼠的治疗作用。方法:SD大鼠随机分为正常对照组,模型组,高、低剂量十二时辰治疗组和阳性对照组。采用大鼠右后足趾皮内注射完全弗氏佐剂制作类风湿性关节炎模型,致炎后采用十二时辰醇提液浸泡涂抹方式给药,模型组及阳性对照组分别给予生理盐水或扶他林涂抹,持续给药28 d。于给药前和给药后每7 d测定足趾容积,给药结束,HE染色检测各组大鼠的组织病理学改变,Elisa方法测定血清IL-1β及RF含量。结果:模型组大鼠在各时间点的足趾肿胀度显著高于正常对照组(P<0.05),血清IL-1β及RF含量显著升高(P<0.01),足趾关节病理破坏明显;高剂量十二时辰醇提液外用可显著降低大鼠的足趾肿胀度(P<0.05),降低血清IL-1β及RF含量(P<0.01),改善关节病理损伤。结论:畲药十二时辰醇提液外用可降低佐剂性关节炎大鼠足肿胀,改善病理损伤,降低血清炎症因子IL-1β及RF含量。Objective To explore the therapeutic effect of external application of the ethanol extract of She medicine“Shi Er Shi Chen”(Clematis florida)on rats with rheumatoid arthritis.Methods SD rats were randomly divided into the normal control group,the model group,the high-dose Clematis florida ethanol extract treatment group,the low-dose Clematis florida ethanol extract treatment group,and the positive control group.Rheumatoid arthritis model was established by intradermal injection of complete Freund's adjuvant into the right hind paw of the rat.After inflammation induction,the rats were treated with ethanol extract of Clematis florida by soaking the paw into the extract.The model group and the positive control group were treated with saline or diclofenac sodium cream,respectively.The rats were continuously administered for 28 days.The hind foot volume was measured before and every 7 days after administration and the paw swelling rate was calculated.HE staining was used to detect the histopathological changes of the foot tissues at the end of the treatment,and Elisa method was used to measure the serum IL-1βand RF levels.Results The paw swelling degree of rats in the model group was significantly higher than that in the normal control group at each time point(P<0.05),and the serum IL-1βand RF levels were significantly increased(P<0.01).HE staining showed that the pathological damage of toe joint was obvious.External application of high-dose Clematis florida ethanol extract could significantly reduce the paw swelling degree of rats(P<0.05),decrease the serum IL-1βand RF levels(P<0.01),and improve the pathology damage of toe joint.Conclusion External arthritis rats,as well as decrease the serum levels of inflammatory factors IL-1βand RF.
