摘  要:甘油醛-3-磷酸脱氢酶(Glyceraldehyde-3-phosphate Dehydrogenase,GAPDH)是糖酵解、糖异生和卡尔文循环的重要酶,同时在植物应对非生物胁迫和激素响应中具有重要作用。为更好地了解NtGAPDH基因家族的特征并预测基因功能,本研究利用生物信息学方法鉴定了烟草GAPDH基因家族,并结合转录组学和qRT-PCR技术分析其在不同条件下的表达模式。结果表明:烟草有18个NtGAPDHs基因,均属于磷酸化GAPDH,分为NtGAPA、NtGAPB、NtGAPC、NtGAPCP 4类,分布在14条染色体上,属于两个亚家族。这些基因编码的氨基酸介于263~542个,等电点在5.12~9.01,相对分子质量为28802.92~59315.82 Da。NtGAPDHs基因的启动子区域含有多种顺式作用元件,包括光响应元件和脱落酸响应元件等。转录组学分析表明,NtGAPDH基因家族成员在不同光环境下的表达模式不同,在光照条件下的表达水平普遍高于黑暗条件,特别是在种子萌发和幼苗早期发育阶段。特定NtGAPDHs基因和蛋白在光照下的高表达暗示其在烟草对光信号响应中的关键作用。研究结果为揭示烟草GAPDH家族基因响应光诱导调控烟草幼苗早期发育的调控机制提供理论参考。Glyceraldehyde-3-phosphate dehydrogenase(GAPDH)is an pivotal enzyme involved in glycolysis,gluconeogenesis,and the Calvin cycle,while also serving a crucial role in plant responses to abiotic stress and hormone signaling.This study aimed to elucidate the characteristics and predict the functions of the NtGAPDH gene family by employing bioinformatics to identify the tobacco GAPDH gene family and investigating their expressionprofiles under dark conditions using a combination of transcriptomics,proteomics,and qRT-PCR.The research revealed that tobacco contained 18 NtGADPHs genes,all of which were classified as phosphorylated GAPDH,and were further divided into four types(NtGAPA,NtGAPB,NtGAPC,NtGAPCP).These genes were located on 14 chromosomes,and were part of two subfamilies.The genes encode amino acids ranging from 263 to 542,exhibiting isoelectric points between 6.54 and Their promoter regions contain various cis-acting elements,including light-responsive and abscisic acid-responsive elements.9.01,and relative molecular masses ranging from 28,802.92 to 59,315.82 Da.The promoter regions of these genes contained various cis-acting elements,including light-responsive and abscisic acid-responsive elements.Transcriptomic and proteomic analyses revealed distinct expression patterns among members of NtGAPDH gene family in response to varying different light conditions.Higher expression levels were observed in light-exposed conditions compared to dark conditions,especially during seed germination and early seedling development stages.The high expression of specific NtGAPDH genes and proteins in response to light indicated their significant involvement in tobacco's light signal response.These findings provide a theoretical basis for understanding the regulatory mechanisms of the tobacco GAPDH gene family in response to light-induced control during early seedling development.
