摘 要:为探究猫尾草(Phleum pratense)种质资源亲缘关系及种子表型差异,本研究采用形态特征测定并结合SSR分子标记对21份国内外猫尾草种质进行研究。结果表明,供试猫尾草种质种子面积、周长、长宽比、长度、宽度和千粒重间存在显著差异。10对SSR分子标记引物共扩增出144条带,多态性条带132条,多态性比率为88.74%。有效等位基因数(Ne)、Nei’s多样性指数(H)和Shannon指数(I)均值分别为1.6266、0.3507和0.5144。材料10676、7629和9038在种子形态聚类和SSR分子聚类时均属于第一类;材料10027和7019属于第二类,且5份材料种子与其他材料相比具有较大较圆、重量较重等优势,两类间遗传相似系数较小,遗传距离较大。本研究筛选出的种质可在新品种选育时作为优势亲本,以增加后代的遗传重组和提高杂种优势。In this research,morphological markers combined with SSR molecular marker were applied to investigate the kinship relationship of germplasm resources and seed phenotypic differences of Timothy(Phleum pratense)from 21 germplasms domestically and foreign.Results of this research demonstrated that there are significant differences between Timothy germplasms on seed area,perimeter,aspect ratio,length,width,and thousand seed weight.144 bands were amplified from 10 pairs of SSR molecular marker primers,in which contained 132 polymorphic bands,resulting a polymorphism ratio of 88.74%.Means of effective number of alleles(Ne),Nei's diversity index(H),and Shannon index(I)were 1.6266,0.3507,and 0.5144,respectively.Material No.10676,7629,and 9038 were clustered as groupⅠfor both seed morphology and SSR molecular.Material No.10027 and 7019 were clustered as groupⅡand the seeds of the five materials were characterized as larger,rounder,heavier compared to other materials.Meanwhile,the genetic coefficient of two cluster groups was relatively small,whereas the genetic distance was high.Germplasm selected from this research can be used as advantage parent in further new cultivar breeding for the aspects of recombination booster and heterosis improvement.
