摘 要:为研究烯效唑(S3307)对自生状态下大豆根瘤菌生长和共生表型影响的机制,比较不同浓度S3307对大豆根瘤菌Sinorhizobium shofinae的细胞生长、显微结构的影响,通过回接试验进行验证,并通过转录组分析揭示其调控机理,最后通过qRT-PCR检测S3307处理对S.shofinae nod基因表达的影响。结果显示:在培养液中添加S3307能够降低S.shofinae细胞的生长速度、改变细胞大小,减少细胞内PHB数量。盆栽回接试验显示S3307处理S.shofinae虽然减少了根瘤数,但提高了大豆植株的茎、叶干重。转录组学分析获得了203个差异基因表达谱,功能富集分析显示这些差异表达基因中被显著抑制的基因主要分布在参与生长以及细胞结构等途径。qRT-PCR分析表明S3307处理抑制了IC52_RS29880(nodA)和IC52_RS31765(nodD)基因的表达,与转录组测序结果一致。试验结果表明植物生长调节剂对纯培养根瘤菌生育存在调控作用,转录组学分析揭示了其调控作用主要发生在细胞过程、定位、新陈代谢、应激响应、生长等过程,这些研究数据有助于我们评价植物调节剂对大豆—根瘤菌共生体系结瘤固氮效率的影响。Aiming to investigate the molecular mechanism of the effect of S3307,a plant growth regulator,on the growth and development of Sinorhizobium shofinae under symbiotic conditions.Various concentrations of S3307 were compared for their effects on cell growth and microstructure of S.shofinae.The effect of S3307 treatment on the transcription level of S.shofinae nod gene was detected by qRT-PCR,which was verified by tieback experiment,and the regulatory mechanism was further clarified by transcriptome analysis.The results indicated that the addition of S3307 to the culture medium slowed down the cell growth rate,induced alterations in the microstructure and reduced the number of PHB of S.shofinae.Tieback test revealed that S3307 treatment reduced the number of nodules but increased the stem and leaf dry weight of soybean plants.Transcriptomic analysis identified 203 differentially expressed genes,with functional enrichment analysis showing that the significantly inhibited genes were mainly involved in pathways related to growth and cell structure.qRT-PCR verification showed that S3307 treatment inhibited the expression of IC52_RS29880(nodA)and IC52_RS31765(nodD)genes,which was consistent with the transcriptome sequencing results.Plant growth regulators have a certain regulatory effect on symbiotic rhizobia and soybeans.The transcriptome results showed that S3307 treatment inhibited the expression of S.shofinae growth gene,and their regulatory effects mainly occurred in cell process,localization,metabolism,stress response and growth.These research findings contribute to our assessment of the impact of plant growth regulators on soil ecological safety.
