摘  要:目的 探讨氧化苦参碱(oxymatrine,OMT)对砷暴露大鼠肝损伤的影响。方法 将64只成年健康清洁级SD雄性大鼠按体重随机分为4组,分别为正常对照(去离子水)组、亚砷酸钠染毒组和低、高剂量OMT干预组,每组16只。采用经口灌胃方式进行亚砷酸钠染毒,每天1次,每周6 d;连续染毒16周后,每组选取8只大鼠,正常对照组和亚砷酸钠用去离子水灌胃,而低、高剂量OMT干预组分别以25、50 mg/kg的OMT溶液灌胃,每天1次,连续4周。分别于染砷16周和OMT干预4周后,检测大鼠血清、尿、肝脏的砷含量和血清中的谷氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)的活力及肝组织中平滑肌肌动蛋白(α-SMA)、透明质酸(HABP1)、Ⅳ型胶原(COL-IV)、转化生长因子-β1(TGF-β1)的浓度以及α-SMA的蛋白表达。结果 砷染毒和OMT干预后,亚砷酸钠染毒组和各剂量OMT干预组大鼠的体重均低于正常对照组;OMT干预后,高剂量OMT干预组大鼠的体重较亚砷酸钠染毒组升高,差异有统计学意义(P<0.05)。砷染毒和OMT干预后,亚砷酸钠染毒组和各剂量OMT干预组大鼠肝、血和尿中的砷含量均高于正常对照组;OMT干预后,高剂量OMT干预组大鼠的肝砷和血砷均低于亚砷酸钠染毒组,各剂量OMT干预组大鼠的尿砷含量均高于亚砷酸钠染毒组,差异有统计学意义(P<0.05)。砷染毒和OMT干预后,亚砷酸钠染毒组和各剂量OMT干预组大鼠血清中ALT和AST活力均高于正常对照组;OMT干预后,各剂量OMT干预组大鼠的ALT活力均低于亚砷酸钠染毒组,高剂量OMT干预组大鼠AST活力低于亚砷酸钠染毒组,差异均有统计学意义(P<0.05)。砷染毒后,亚砷酸钠染毒组和各剂量OMT干预组大鼠肝组织HABP1、COL-IV、α-SMA的浓度均高于正常对照组;OMT干预后,各剂量OMT干预组大鼠肝组织COL-IV和α-SMA的浓度均低于亚砷酸钠染毒组,且高剂量OMT干预组大鼠肝组织HABP1的浓度低于亚砷酸钠染毒组,�Objective To understand the protective effect of oxymatrine(OMT)on liver injury in rats with chronic arsenic exposure.Methods Sixty-four adult male SD rats were randomly divided into 4 groups,16 in each,the control group(deionized water),the sodium arsenite exposed group and the low and high dose OMT intervention groups.Sodium arsenite was exposed by oral gavage,once a day,6 days a week.After 16 weeks of continuous exposure,8 rats were chosen from each group,the control group and sodium arsenite were treated with deionized water,and the low-dose and high-dose OMT intervention groups were intragastrically administered with 25 and 50 mg/kg OMT solution,once a day for 4 consecutive weeks.After 16 weeks of arsenic exposure and 4 weeks ofOMT intervention,the levels of arsenic in serum,urine and liver and the activities of glutamate aminotransferase(ALT)and aspartate aminotransferase(AST)in the serum and the expression levels of smooth muscle actin(α-SMA),hyaluronic acid(HABP1),typeⅣcollagen(COL-Ⅳ),transforming growth factor-β1(TGF-β1)andα-SMA protein in liver tissues were determined.Results The body weight of the rats in the sodium arsenite exposed group and the OMT intervention group of each dose was lower than that of the control group.The weight of the rats in the high-dose OMT intervention group was significantly higher than that of sodium arsenite(P<0.05).The levels of arsenic in the liver,blood and urine in the sodium arsenite exposed group and the OMT intervention group were higher compared with the control group.In the high-dose OMT intervention group,the liver arsenic and blood arsenic were lower than the sodium arsenite-exposed group,and the urine arsenic content of rats in each dose of OMT intervention group was higher than that of the sodium arsenite-exposed group,the difference was statistically significant(P<0.05).After arsenic exposure and OMT intervention,the serum ALT and AST activities in the sodium arsenite exposure group and the OMT intervention group of each dose were higher than those in
