摘  要:痛风性关节炎是由尿酸钠晶体沉积在关节和非关节组织引起的一种常见的晶体相关性疾病,可导致炎症和关节损伤,主要跟遗传、环境和代谢因素密切相关。它的发生通常与血清尿酸水平升高显著相关,在持续高尿酸血症的情况下,尿酸钠晶体在关节和关节周围间隙的慢性沉积以及随后先天免疫系统的激活是痛风性关节炎发生的核心机制。其中NOD样受体家族蛋白3炎症小体及一些细胞因子在痛风性关节炎激活过程中的确切分子机制尚未阐明,痛风性关节炎反复发作导致痛风石形成的治疗方案有限。因此,寻找有效的药物作用靶点靶向治疗痛风性关节炎显得尤为重要。Gouty arthritis is a common crystal-related disease caused by the deposition of sodium urate crystals in articular and non-articular tissues,which can lead to inflammation and joint damage.It is mainly closely related to genetic,environmental and metabolic factors.Its occurrence is usually significantly associated with elevated serum uric acid levels.In the presence of persistent hyperuricemia,chronic deposition of sodium urate crystals in the joint and periarticular space and subsequent activation of the innate immune system are central mechanisms in the development of gouty arthritis.Among them,the exact molecular mechanism of NOD-like receptor family protein 3 inflammasome and some cytokines in the activation of gouty arthritis has not been elucidated.The treatment of recurrent gouty arthritis leading to the formation of tophus is limited.Therefore,it is particularly important to find effective drug targets for the treatment of gouty arthritis.
