摘 要:目的:观察艾烟对APP/PS1小鼠认知能力及下丘脑-垂体-性腺轴激素水平的影响。方法:27只雄性APP/PS1小鼠随机分为模型组、迷迭香精油组(以下简称精油组)、艾燃烧生成物组(以下简称艾烟组),每组9只。9只相同遗传背景的C57BL/6小鼠为空白组。精油组和艾烟组小鼠分别采用浓度为1%的迷迭香精油及浓度为10~15 mg/m3的艾烟进行吸嗅干预,空白组和模型组小鼠仅抓取固定。12周后用Morris水迷宫实验检测小鼠的学习记忆能力,ELISA法检测血清促性腺激素释放激素(GnRH)、促黄体生成素(LH)、促卵泡刺激素(FSH)以及睾酮(T)水平,刚果红染色法观察小鼠前额叶皮层β样淀粉蛋白(Aβ)的沉积情况。结果:与模型组比较,精油组和艾烟组逃避潜伏期分别在第3~5天和第4~5天显著缩短(P<0.05),穿越平台次数、平台区域游泳时间均显著增加(P<0.05),LH、FSH含量显著降低(P<0.05,P<0.01),T含量显著升高(P<0.05),前额叶皮层Aβ沉积减少;精油组平台区域游泳路程占比显著增加(P<0.05),GnRH含量显著降低(P<0.01)。结论:艾烟可以通过纠正HPG轴激素的紊乱,有效减少APP/PS1小鼠脑内Aβ沉积,改善认知能力。Objective:To observe the effects of moxa smoke on cognitive ability and hypothalamic-pituitary-gonadal axis hormone levels in APP/PS1 mice.Methods:Twenty-seven male APP/PS1 mice were randomly divided into model group,rosemary essential oil group(hereinafter referred to as essential oil group)and moxa combustion products group(hereinafter referred to as moxa smoke group),with nine mice in each group.Nine C57BL/6 mice with the same genetic background were used as the blank group.The essential oil group and the moxa smoke group were respectively treated with 1%rosemary essential oil and 10-15 mg/m'moxa smoke for sniffing intervention.Mice in the blank group and the model group were only grasped and fixed.After 12 weeks,the Morris water maze test was used to detect the learning and memory ability of the mice.ELISA was used to detect serum levels of gonadotropin releasing hormone(GnRH),luteinizing hormone(LH),follicle stimulating hormone(FSH)and testosterone(T).Congo red staining was used to observe the deposition ofβ-amyloid protein(Aβ)in the prefrontal cortex of mice.Results:Compared with model group,the escape latency of essential oil group on days 3-5 and moxa smoke group on days 4-5 shortened significantly(P<0.05).The number of crossing the platform and swimming time in the platform area were significantly increased(P<0.05).The contents of LH and FSH were significantly decreased(P<0.05,P<0.01),T content significantly increased(P<0.05).Aβdeposition was reduced in the prefrontal cortex;The proportion of swimming distance in the platform area of the essential oil group was significantly increased(P<0.05),and the GnRH content was significantly decreased(P<0.01).Conclusion:Moxa smoke can effectively reduce the deposition of Aβin the brain of APP/PS1 mice and improve the cognitive ability by correcting the disorder of HPG axis hormones.
