摘　　要：目的:从坎离颗粒中筛选出抗慢性心力衰竭主效应中药,再验证筛选方的疗效。方法:采用腹主动脉缩窄诱导慢性心力衰竭大鼠模型,运用DPSVT.05版软件产生U20(2010)表,采用拟水平法确定均匀设计实验方案对坎离颗粒进行10因子(10味中药)10水平(10种剂量)20次实验。以血流动力学、心肌肌质网钙摄取速率作为筛选指标,运用均匀设计方法进行逐步回归分析,筛选出主效应中药。再次运用该模型进行验证实验,对慢性心力衰竭模型进行心脏功能学、血流动力学、心肌肌质网钙摄取速率检测,对所得中药筛选方的抗慢性心力衰竭疗效进行验证。结果:坎离颗粒中黄芪、附子、莪术、麦冬且生药剂量分别为5.4、1.8、1.8、1.8 g/kg改善慢性心力衰竭疗效最佳,验证实验显示筛选结果准确、可靠。结论:黄芪、附子、莪术、麦冬是坎离颗粒改善慢性心力衰竭的主效应中药。Objective:Screening the main effect traditional Chinese medicine ingredient of anti-heart failure effects from the Kanli Granules,and then verify the efficacy of the filtering party.Methods:Abdominal aortic coarctation surgery was used to induce chronic heart failure of mice models,and DPSVT.05 data processing system was used to generate the U20(2010)table to group experiment for 10 fiavor of Kanli Granules.Based on blood fow dynamics and muscle mesh calcium activity as screening indicators,a uniform design method was used to gradually return and analyze,so as to screen the main effect of traditional Chinese medicine.The model was used again for verification experiments,and the heart failure model was used to conduct a heart-colored Doppler,blood fow dynamic,and muscle mesh calcium activity testing,and verify the effect of the screened anti-chronic heart failure Chinese medicine.Results:When the dried medicinal herbs dosage of Astragali Radix,Aconiti Lateralis Radix Praeparate,Curcumae Rhizoma and Ophiopogonis Radix in Kanli Granules were 5.4,1.8,1.8,1.8 g/kg,respectively,the effect for improving chronic heart failure was the best.The screening results were accurate and reliable.Conclusion:Astragali Radix,Aconiti Lateralis Radix Praeparate,Curcumae Rhizoma and Ophiopogonis Radix are the main effect Chinese medicine in Kanli Granules to improvechronicheartfailure.
