猕猴骨关节炎阳虚络痹病证结合模型的构建与评价

摘　　要：目的:构建猕猴骨关节炎(OA)阳虚络痹病证结合动物模型。方法:健康猕猴8只随机分为实验组、对照组,每组4只,对实验组进行肌注醋酸泼尼松龙建立肾阳虚证模型,以手术方法建立OA模型。观察指标包括一般情况、体征、血液生化指标、关节磁共振成像检查,肾脏切片。结果:与对照组比较,实验组猕猴毛色干枯,并逐渐脱落,精神萎靡,活动减少,并逐渐出现纳差、小便清长等阳虚体征,肾脏病理切片出现淋巴细胞浸润。对照组猕猴舌淡红苔薄白,实验组不同程度的舌质淡偏白。实验组睾酮、促肾上腺皮质激素、四碘甲状原氨酸显著低于对照组(P<0.01)。磁共振成像结果显示猕猴关节出现了软骨侵蚀损伤,滑膜增厚、水肿,骨髓水肿。结论:本研究通过醋酸泼尼松龙联合手术的方法初步构建了猕猴OA阳虚络痹的病证结合模型。Objective:To develop a kind of combined animal model of yang deficiency collateral bi syndrome and osteoarthritis(OA)in rhesus monkeys.Methods:Eight healthy rhesus monkeys were randomly assigned to an experimental or a control group,with four monkeys in each group.The experimental group received intramuscular injections of prednisolone acetate to induce yang deficiency syndrome,while OA was induced through surgical intervention.Evaluated parameters included physical signs,blood biochemical markers,joint imaging via magnetic resonance imaging(MRI),and kidney sections.Results:Compared to the control group,monkeys in the experimental group exhibited symptoms such as dry and sheding fur,lethargy,reduced activity,poor appetite,and frequent urination,consistent with yang deficiency.Pathological kidney examination revealed lymphocyte infiltration in the experimental group.Tongue examination showed a pale tongue with varying degrees of thin white coating in the experimental group,compared to a pale red tongue with a thin white coating in the control group.Testosterone,adrenocorticotropic hormone,and thyroxine levels were significantly lower in the experimental group than in the control group(P<0.01).MRI results revealed that the monkey's joint exhibited cartilage erosion,synovial thickening,edema,and bone marrow edema.Conclusion:The combination of prednisolone acetate administration and surgical intervention successfully establishes a preliminary model of OA with yang deficiency collateral bi syndrome in rhesus monkeys.
