摘  要:本研究对墨兰APETALA1-like(AP1)基因的克隆、分析和实时荧光定量(RT-qPCR)表达发现,所克隆的AP1为MIKC型MADS-box基因,具有保守的MADS区和相对保守的K区,且与朵丽蝶兰(AFD36438.1)同源性最高,为100%。将此基因命名为CsAP1,登录号为MW654192。RT-qPCR实验分析显示,CsAP1在墨兰各个时期的组织器官中均有表达,相对表达丰度从高到低依次为:花萼(Ⅲ)>叶片(Ⅲ)>根(Ⅰ)>唇瓣(Ⅲ)>花葶(Ⅲ)>叶(Ⅱ)>根(Ⅲ)>根(Ⅱ)>花葶(Ⅱ)>花瓣(Ⅲ)>叶(Ⅰ)>花蕾(Ⅱ)>子房(Ⅲ)>合蕊柱(Ⅲ)。盛花期花萼和叶片中表达丰度特别高,该基因参与花器官萼片的形成,并可能调节开花时间。从生长发育时期上看,盛花期的表达量最高,说明CsAP1调控开花时间。对于叶,3个时期表达量逐渐升高,推断CsAP1促进开花。对于生殖器官花来说,该基因的表达量从花萼到花瓣再到合蕊柱自外而内逐渐降低。本研究为进一步研究墨兰AP1基因的功能和分子机制提供了基础。In this research,the APETALA1-like(AP1)gene of Cymbidium sinense was cloned,analyzed and expressed by real-time fluorescence quantitative analysis(RT-qPCR).The results showed that the cloned AP1 was MIKC MADS-box gene with conserved MADS region and relatively conserved K region,and had the highest homology(100%)with Phalaenopsis hybrid(AFD36438.1).The gene was named as CsAP1,and the accession number was MW654192.RT-qPCR analysis showed that CsAP1 was expressed in tissues of Cymbidium sinense in dif ferent periods,and the order of relative expression abundance from high to low was calyx(Ⅲ)>leaf(Ⅲ)>root(Ⅰ)>lip petal(Ⅲ)>inflorescence rachis(Ⅲ)>leaf(Ⅱ)>root(Ⅲ)>root(Ⅱ)>inflorescence rachis(Ⅱ)>petal(Ⅲ)>leaf(Ⅰ)>bud(Ⅱ)>ovary(Ⅲ)>gynostemium(Ⅲ).CsAP1 was highly expressed in calyx and leaves at full-blossom stage,which was involved in the formation of sepals of flower organs and might regulate flowering time.In terms of growth and development period,the expression level at full-blossom stage was the highest,which indicated that CsAP1 regulated flowering time.With respect to leaves,the expression level increased gradually in three stages,which suggested that CsAP1 promoted flowering.For flowers,the expression level of CsAP1 gradually decreased from calyx to petal and then to gynostemium.This research provides a basis for further studying the function and molecular mechanism of AP1 gene in Cymbidium sinense.
