摘 要:野大麦是优质生态改良草种,但目前利用分子标记开发方面研究较少。本研究在野大麦的214676条unigene序列中查找SSR位点,共检测到24877个SSR分布于21618条unigene序列中。重复类型数量由多到少的顺序为单核苷酸>三核苷酸>二核苷酸。对野大麦SSR不同重复类型在不同重复次数下的数量进行统计分析,结果表明,重复次数变化范围在5~24次之间,占比65.70%,说明基于短芒大麦转录组SSR开发具有很大可能性。重复类型中,SSR 5次重复的数量最多,为5237个,是总数的21.05%;其次为10次重复,共4861个,占总数的19.54%。利用Primer 3软件设计SSR引物,unigene序列上的24877个SSR位点中设计成功的引物共有20802个,成功率为83.62%。对引物进行随机检测,成功率为80%。本研究为野大麦SSR分子标记开发应用、种质资源遗传性评价、分子标记的辅助育种等提供参考。Hordeum brevisubulatum is a high-quality ecologically improved grass species,but currently there is little research on the development of molecular markers.Searching for SSR loci in 214676 unigene sequences of H.brevisubulatum,a total of 24877 SSRs were detected in 21618 unigene sequences.The order of the number of repeat types was mononucleotide>trinucleotide>dinucleotide.The statistical analysis of the number of different repeat types of H.brevisubulatum under different repeat times showed that the variation range of the repeat times was between 5 and 24 times,accounting for 65.70%,indicating that the development of SSR based on the transcriptome of H.brevisubulatum has a great possibility.Among the SSR repeat types,the number of SSRs with 5repetitions was the largest,reaching 5237,accounting for 21.05%of the total;followed by 10 repetitions,with a total of 4861,accounting for 19.54%of the total.SSR primers were designed using Primer 3 software,20802 primers were successfully designed out of the 24877 SSR loci in the unigene sequence,with a success rate of 83.62%.Random detection of primers had a success rate of 80%.This study provides references for the development and application of SSR molecular markers of H.brevisubulatum,genetic evaluation of germplasm resources,and assisted breeding of molecular markers.
