摘 要:为了解金银花及近缘种之间的遗传背景差异与亲缘关系远近,本研究利用SSR分子标记技术对35份药用金银花种质资源进行遗传多样性分析和指纹图谱构建。结果表明,所选用的23对引物有效等位基因数和期望杂合度的平均值分别为2.891和0.618,Shannon信息指数和Nei's遗传多样性指数的平均值分别为1.300和0.609,各位点的多态性信息含量平均值为0.597,表现出较高的多态性。在遗传相似系数为0.250处,可将35份供试材料分为4类。本研究运用核心引物法构建了金银花及其近缘种的指纹图谱,实现了从分子水平上对金银花种质资源的快速鉴别。本研究为区分金银花真伪品、丰富优良性状和选育优质品种提供了科学依据。In order to understand the differences of genetic background and genetic relationship between Lonicera japonica germplasm resources,in this study,genetic diversity analysis and fingerprint construction of 35 medicinal Lonicera japonica germplasm resources were carried out by SSR molecular marker technology.The results showed that the average number of effective alleles and expected heterozygosity of 23 pairs of primers were 2.891 and0.618,respectively.The average Shannon information index and Nei's genetic diversity index were 1.300 and0.609,respectively.The mean value of polymorphism information content of each site was 0.597,showing a high polymorphism.At the genetic similarity coefficient of 0.250,35 tested materials were divided into four categories.In addition,the fingerprint of Lonicera japonica germplasm resources was constructed by core primer method,and the rapid identification of Lonicera japonica and the related species at the molecular level was realized.This study provides a scientific basis for distinguishing genuine and fake Lonicera japonica,enriching excellent characters and breeding high-quality varieties.
