摘  要:本研究以巴马火麻为试验材料,采用SRAP分子标记技术,筛选与巴马火麻性别相关或连锁的特异条带。结果显示,引物ISSR839在巴马火麻雌雄单株之间存在较好多态性且条带清晰。因此,利用ISSR839引物检测雌雄单株之间的遗传稳定性,用于构建极端雌雄DNA池。此外,560对SRAP引物组合中24对引物在雌雄DNA池间能扩增出差异条带,其中引物对me11/em12在雄性DNA池中能稳定的扩增出一条1431 bp的特异条带,而相应的雌性DNA池未能扩增出此条带。为了将此特异条带转化为稳定的SCAR标记,基于me11/em12引物对的基础,分别向5'和3'端延伸了2~3个碱基,获得SCAR标记的特异引物HXF/HXR,该引物经火麻雌雄单株验证。结果显示,雄性单株能扩增出唯一的1436 bp条带,命名为SCAR-1436,而相应的雌性单株未能扩增此条带。该特异片段经克隆测序与比对,在NCBI上未找到与该片段同源的序列。本研究结果可为苗期的火麻植株进行雌雄性别鉴定,合理控制雌雄植株比例,提高火麻籽产量提供技术手段。In this study,Bama hemp was used as experimental material,the specific fragments linked or related to hemp sex traits were screened using sequence-related amplified polymorphism(SRAP)molecular marker technology.The results showed that the primer ISSR839 had abundant polymorphisms and clear bands in hemp male and female individuals,then the primer ISSR839 was used to detect the genetic stability between male and female individual plants and to construct extreme male and female DNA pools.In addition,among 560 pairs of SRAP primers,24 pairs of primers could amplify different bands between male and female DNA pools,among which,the primer pair me11/em12 could steadily amplify a specific fragment of 1431 bp from the male DNA pool,while this fragment could not be amplified in the corresponding female DNA pool.To convert this specific band into a stable SCAR(sequence characterized amplified region)marker,2~3 bases were extended at the 5'and 3'ends based on the me11/em12 primer pair,resulting in the specific SCAR primer HXF/HXR.This primer was validated in male and female single plants of hemp.The results showed that a unique 1436 bp band could be amplified in male plants,named SCAR-1436,while the corresponding female plants did not amplify this band.Cloning,sequencing,and comparison of this specific fragment revealed no homologous sequences in the NCBI database.The results of this study can provide a technical means for the sex identification of hemp plants at the seedling stage,reasonably control the ratio of male to female plants,and improve hemp seed yield.
