摘  要:对野外采集的皱木耳开展真菌种属(internal transcribed spacer,ITS)测序及构建系统发育树鉴定菌株分支,并研究其菌丝最适温度、pH值、无机盐浓度比及培养基最优配方等生物学特性。结果表明,检测序列与皱木耳Au-ricularia delicata支持率达99%;菌丝培养最宜温度为25℃,最宜pH值为6,无机盐PO43-和Mg2+浓度比为1.0%∶1%,正交结果显示最优配方为糊精粉10.0 g、乳糖5.0 g、牛肉膏2.0 g、酵母浸粉2.0 g、KH2PO41 g、MgSO4·7H2O 1 g、维生素B110 mg、琼脂20 g、去离子水1000 mL。The internal transcribed spacer(ITS)sequencing and phylogenetic analysis were carried out to identify the Auricularia delicata samples collected in the field.The optimal temperature,pH value,inorganic salt concentration ratio,and medium formula for the mycelial growth were determined.The results showed that the sequence identity between the collected samples and A.delicata was 99%.The optimal culture conditions for mycelial growth were 25℃,pH6,and PO43-:Mg2+concentration ratio of 1.0%:1%.The orthogonal test re-sults showed that the optimal formula was composed of 10.0 g dextrin powder,5.0 g lactose,2.0 g beef extract,2.0 g yeast extract,1 g KH2PO4,1 g MgSO4·7H2O,10 mg vitamin B1,20 g agar,and 1000 mL deionized wa-ter.
