摘 要:目的:优化仙人掌胃康胶囊的含量测定方法,以提升仙人掌胃康胶囊含量测定的方便性和科学性。方法:采用加热回流提取样品,高效液相色谱法测定样品中橙皮苷含量;色谱条件:色谱柱为Agilent TC-C18柱(250 mm×4.6 mm,5μm),流动相采用乙腈-0.1%磷酸溶液(20∶80),柱温为室温,PDA检测器,检测波长283 nm,流速1.0 mL·min^(-1)。结果:HPLC方法学考察表明,橙皮苷在2.0742~414.8496μg/mL范围内线性关系良好(R^(2)=0.9997),精密度RSD≤0.56%,重复性RSD≤1.12%(n=6),稳定性RSD≤2.03%,供试品在以25 mL甲醇为提取溶剂,加热时间30 min时平均回收率为94.60%,RSD=2.31%(n=6)。结论:优化HPLC法测定仙人掌胃康胶囊中橙皮苷的含量,样品提取方法简便,结果准确,重复性好,可用于仙人掌胃康胶囊中橙皮苷含量的测定,为更好地控制仙人掌胃康胶囊质量稳定和可靠提供科学依据。We optimized the content determination method for Cactus Weikang Capsules,so as to improve the convenience and scientificity of the method.The sample was extracted by heating reflux,and the content of hesperidin in the sample was determined by HPLC.Chromatographic conditions;the chromatographic column was Agilent TC-C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid solution(20∶80),the column temperature was room temperature,the PDA detector was adopted,the detection wavelength was 283 nm,and the flow rate was 1.0 mL/min.The results showed that hesperidin had good linear relationship in the range of 2.0742μg/mL to 414.8496μg/mL(R^(2)=0.9997);the precision RSD≤0.56%,the repeatability RSD≤1.12%(n=6),and the stability RSD≤2.03%;the average recovery rate of the sample was 94.60%,RSD=2.31%(n=6)when heated for 30 minutes with 25 mL methanol as the extraction solvent.The optimized HPLC method is simple,accurate and reproducible,and can be used for the determination of hesperidin in Cactus Weikang Capsules.This study has provided scientific basis for the quality control of Cactus Weikang Capsules.
