摘  要:目的 建立高效液相色谱法(HPLC)测定宫炎康颗粒中菊苣酸的含量。方法 以Agilent 5 TC-C18(4.6 mm×250 mm,5.0μm)为色谱柱,流动相为乙腈-0.1%甲酸溶液(含0.1%三乙胺)(梯度洗脱),检测波长为328 nm,流速为1 ml/min,柱温35℃,进样量10μl。结果 菊苣酸在2.09~104.49μg/ml浓度范围内线性关系良好(R^(2)=0.9992),平均加样回收率为103.84%,精密度、重复性、稳定性的RSD<2.0%。24批样品菊苣酸的含量范围为66.76~364.85μg/g。结论 所建方法能有效测定宫炎康颗粒中菊苣酸的含量,可为完善该制剂的质量标准提供借鉴。Objective To establish a high performance liquid chromatography(HPLC) method for the determination of chicoric acid in Gongyankang granules.Methods The determination was performed on Agilent 5 TC-C18(4.6 mm×5.0×250 mm) column with mobile phase consisting of acetonitrile-0.1% formic acid solution(Contains 0.1% triethylamine)(gradient elution).The detection wavelength was set at 328 nm,the flow rate was 1 ml/min,the column temperature was 35 ℃,and the sample size was 10 μl.Results The linear relationship of chicoric acid was good in the concentration range of 2.09~104.39 μg/ml(R~2=0.9992),the average recovery rate was 103.84%,and the RSD of precision,repeatability and stability was less than 2.0%.The content of chicoric acid in 24 batches of samples ranged from 66.76 to 364.85 μg/g.Conclusion The established method can effectively determine the content of chicoric acid in Gongyankang granules,and can provide reference for improving the quality standard of this preparation.
