摘 要:目的:优化复合酶辅助提取流苏香竹叶黄酮工艺条件,评价流苏香竹叶黄酮体外抗糖尿病潜力。方法:采用响应面法优化流苏香竹叶黄酮超声协同复合酶提取工艺,以提取物及其萃取组分的总黄酮得率为评价指标,研究其对α-葡萄糖苷酶和醛糖还原酶的抑制活性。结果:流苏香竹叶黄酮提取的最佳工艺条件为料液比1∶24 g/mL,复合酶比例(果胶酶∶纤维素酶)为1∶3,酶解温度51℃,pH值为6.0,此条件下黄酮得率为8.95%。流苏香竹叶粗提物水相萃取组分的总黄酮含量为1.175 mg/mL。生物活性研究表明,与阿卡波糖相比,乙酸乙酯和二氯甲烷相萃取组分的α-葡萄糖苷酶抑制活性更高,分别为阿卡波糖的4.14和9.70倍。同时,二氯甲烷相萃取组分有最强的醛糖还原酶抑制活性(IC_(50)=0.167 mg/mL),表明该溶剂能更好的从流苏香竹叶中富集这两种酶活性抑制的有效成分。皮尔森相关性分析表明,黄酮类化合物是流苏香竹叶中主要的活性成分,与α-葡萄糖苷酶和醛糖还原酶抑制活性呈中高度相关。结论:流苏香竹叶黄酮在抗糖尿病药物或保健产品等领域的开发潜力较大。Objective:To optimize the extraction conditions of flavonoids from the leaves of Chimonocalamus fimbriatus with complex enzyme assistance and evaluate their potential for in vitro anti-diabetes activity.Methods:The response surface methodology was used to optimize the ultrasonic-coordinated complex enzyme extraction technology of flavonoids from the leaves.The total flavonoid yield of the extract and its extract components were used as the evaluation index,and the inhibitory activities of flavonoids on α-glucosidase and aldose reductase were studied.Results:The optimal extraction conditions were as follows:solid-liquid ratio 1∶24,ratio of the complex enzyme(pectinase∶cellulase)1∶3,hydrolysis temperature 51℃,and pH 6.0.Under the optimal conditions,the yield of flavonoids was 8.95%.The total flavonoid content of the aqueous phase extraction component of the crude extract was 1.175 mg/mL.The bioactivity studies showed that theα-glucosidase inhibitory activity of ethyl acetate extract and dichloromethane extract was 4.14 and 9.70 times higher than that of acarbose.At the same time,the dichloromethane extract had the strongest aldose reductase inhibitory activity(IC_(50)=0.167 mg/mL),indicating that this solvent could better enrich the active components of these two enzymes in the leaves.Pearson correlation analysis showed that flavonoids were the main active components in the leaves,and the inhibitory activities ofα-glucosidase and aldose reductase were highly correlated.Conclusion:The flavonoids of C.fimbriatus leaves have great potential in anti-diabetes drugs or health products.
