摘  要:研究通过比较红光和白光条件下京海黄鸡下丘脑的转录组测序,筛选红光影响下丘脑生殖激素分泌的关键基因。试验将300只16周龄京海黄鸡母鸡随机分为2组,饲养于红光(660 nm)和白光(400~760 nm)条件下,每组随机选择3只京海黄鸡,采集下丘脑组织,进行转录组测序和分析。结果显示:与白光组相比,红光组共获得172个差异表达基因(DEGs),其中63个DEGs上调,109个DEGs下调;结合GO和KEGG数据库对DEGs进行功能注释和通路分析显示,DEGs显著富集到神经活性配体一受体相互作用、酪氨酸代谢、钙离子信号通路和间隙连接通路等信号通路;RT-qPCR对随机挑选的6个基因验证结果与测序结果一致。结果表明,TPH2、VAX1、EOMES、CFTR和STRA6 5个差异基因与下丘脑生殖激素分泌相关,其可能在红光影响京海黄鸡下丘脑生殖激素分泌中发挥重要作用。In this study,transcriptome sequencing was used to analyze the hypothalamus of Jinghai yellow chicken under red light(RL)and white light(WL)and screen the key genes of red light affecting the secretion of reproductive hormone in hypothalamus.Three hundred 16-week-age Jinghai yellow chickens were divided into 2 groups randomly and raised under red light(660 nm)and white light(400 to 760 nm),respectively.Three Jinghai yellow chickens were selected randomly from each group and the hypothalamus were collected for transcriptome sequencing and analysis.The results showed that a total of 172 differentially expressed genes(DEGs)were obtained from RL group compared with WL group,of which 63 DEGs were up-regulated and 109 DEGs were down-regulated.GO and KEGG analysis showed that DEGs were significantly enriched in neuroactive receptor interactions,tyrosine metabolism,calcium signaling pathway and gap junction.Six DEGs were selected randomly and verified by RT-qPCR and the results were consistent with the sequencing results.The results indicated that TPH2,VAX1,EOMES,CFTR and STRA6 were involved in secretion of reproductive hormone in hypothalamus,which might play important role in red light affecting the secretion of reproductive hormone of hypothalamus in Jinghai yellow chicken.
