摘 要:为分析信阳地区鹅星状病毒(GAstV)的遗传变异情况,研究从信阳某鹅场痛风病鹅采集病料,进行病原分离鉴定和全基因组扩增测序,获得GAstV基因组,随后进行遗传进化和同源性分析。结果显示:分离一株GAstV,命名为HNxy2111,基因组全长7 175 bp;HNxy2111基因组及ORF2基因均与参考毒株G519亲缘关系最近,处于同一分支,属于GAstV-Ⅱ型;HNxy2111基因组、ORF1a、ORF1b编码氨基酸序列与GAstV-Ⅱ型中的G519同源性最高,均为99.2%。ORF1b编码的氨基酸序列与GD AHAU2同源性最高,为100%。与G519相比,HNxy2111 ORF1a、ORF1b和ORF2编码的蛋白质分别发生了8处、3处和5处突变。综上所述,HNxy2111属于GAstV-Ⅱ型,与国内经典鹅星状病毒有较近的遗传关系,ORF1a、ORF1b和ORF2编码的蛋白有一定的突变。In order to analyze the genetic variation of goose astrovirus(GAstV)in Xinyang area,this study collected disease samples from gout-disease geese in a goose farm in Xinyang area,conducted virus isolation,identification and whole genome amplification sequencing,obtained GAstV genome,and then conducted genetic evolution and homology analysis.The results showed that a strain of GAstV named HNxy2111 with a total genome length of 7175 bp was isolated.The HNxy2111 genome and ORF2 gene were closely related to the reference strain G519,and belong to GAstV-Ⅱtype.HNxy2111 genome,ORF1a and ORF1b amino acid sequences had the highest homology with G519 in GAstV-Ⅱtype,all of which was 99.2%.The amino acid encoded by ORF1b had the highest homology with GD AHAU2,reaching 100%.Compared with G519,HNxy2111 had 8,3 and 5 mutations in the proteins encoded by ORF1a,ORF1b and ORF2,respectively.In summary,HNxy2111 belonged to GAstV-Ⅱtype and had a close genetic relationship with the domestic classic goose astrovirus,with certain mutations in the proteins encoded by ORF1a,ORF1b and ORF2.
