外源ACC对龙须菜(Gracilariopsis lemaneiformis)果孢子放散的生理影响及转录组分析

摘  要:龙须菜(Gracilariopsis lemaneiformis)是我国产量第二大的养殖海藻,以龙须菜为材料,分别添加5种浓度的1-氨基环丙烷-1-羧酸(ACC),探究了其对龙须菜果孢子放散的作用,结果发现250μmol/L的ACC促进果孢子放散效果最佳。外源添加250μmol/L的ACC后,第2天和第5天龙须菜光合作用进一步下降,第5天后超氧化物歧化酶(SOD)、过氧化物酶(POD)、谷胱甘肽还原酶(GR)活性以及还原型抗坏血酸(AsA)含量显著上升,表明龙须菜果孢子的放散还和抗氧化系统密切相关。转录组结果显示ACC处理后第5天光合作用相关基因表达量下调,糖酵解通量增加,TCA循环和氮代谢途径的基因表达量显著上调;外源ACC显著上调了茉莉酸合成途径相关基因的表达,但对乙烯合成途径相关基因无显著影响。研究结果验证了ACC促进龙须菜果孢子放散的作用,为龙须菜的快速繁育提供了新的技术支撑。Gracilariopsis lemaneiformis is the second largest mass culture seaweed in China.In this study,we used G.lemaneiformis as the material and investigated the effects of five different concentrations of 1-aminocyclopropane-1-carboxylic acid(ACC)on the carpospore release of G.lemaneiformis.It was observed that 250μmol/L of ACC showed the most optimal effect in promoting spore release.Following the exogenous addition of 250μmol/L ACC,the photosynthesis of G.lemaneiformis further declined on the second and fifth days.Additionally,spore release in G.lemaneiformis was closely associated with the antioxidant system.By the fifth day after ACC treatment,the activities of superoxide dismutase(SOD),peroxidase(POD),and glutathione reductase(GR),as well as the content of reduced ascorbic acid(AsA),significantly increased.Moreover,transcriptomic analysis revealed that on the fifth day after ACC treatment,the expression levels of genes associated with the photosynthetic pathway were downregulated.Conversely,genes related to glycolysis,tricarboxylic acid(TCA)cycle,and nitrogen metabolism pathways were significantly upregulated.Finally,exogenous ACC significantly upregulated genes related to jasmonic acid synthesis pathway,while showing no significant effect on genes related to the ethylene synthesis pathway.This study demonstrated the role of ACC in promoting the release of carpospores in G.lemaneiformis,providing new techniques for the germplasm breeding of G.lemaneiformis.
