摘 要:为揭示白藜芦醇(RES)是否通过介导过氧化物酶体增殖物激活受体-γ共激活因子-1α(PGC-1α)起到缓解奶牛乳腺上皮细胞(bMECs)炎性反应,本试验对PGC-lα进行干扰(si-PGC-lα),采用单因素随机试验设计方法,设置si-NC+LPS组(含10μg/mL LPS配制的饥饿培养基)、si-PGC-lα+LPS组(si-PGC-lα干扰片段+含10μg/mL LPS配制的饥饿培养基)、si-NC+RES+LPS组(含10μg/mL LPS以及15μmol/L RES共同配制的饥饿培养基)、si-PGC-lα+RES+LPS组(si-PGC-lα干扰片段+10μg/mL LPS以及15μmol/L RES共同配制的饥饿培养基)。结果发现:si-PGC-lα后加入LPS可显著或极显著上调炎性因子白细胞介素-6(IL-6)和白细胞介素-8(IL-8)的mRNA相对表达水平和含量(P<0.05或P<0.01),极显著上调氧化因子丙二醛(MDA)的mRNA相对表达水平(P<0.01),显著或极显著上调促凋亡因子B-细胞淋巴瘤-2相关X蛋白(BAX)的mRNA相对表达水平和bMECs凋亡率(P<0.05或P<0.01),极显著降低抗氧化基因谷胱甘肽过氧化物酶(GSH-Px)的mRNA相对表达水平(P<0.01);而RES可逆转这一反应。当si-PGC-lα后再用RES处理则无法缓解LPS诱导的bMECs产生的氧化应激、炎性反应和细胞凋亡,说明RES是通过激活PGC-1α起到缓解LPS诱导的bMECs炎性损伤和细胞凋亡。由此可见,外源添加RES可通过激活PGC-1α起到缓解LPS诱导的bMECs线粒体损伤、氧化应激、炎性反应并抑制细胞凋亡。In order to reveal whether resveratrol(RES)alleviated inflammatory reaction of bovine mammary epithelial cells(bMECs)by mediating peroxisome proliferator-activated receptor-gamma coactivator-1α(PGC-1α),this present experiment was performed on the PGC-lαwas interfered(si-PGC-lα)by using a one-way randomized trial design method,setting up si-NC+LPS group(hunger medium prepared with 10μg/mL LPS),si-PGC-lα+LPS group(si-PGC-lαinterference fragment+hunger medium prepared with 10μg/mL LPS),si-NC+RES+LPS group(hunger medium co-prepared with 10μg/mL LPS and 15μmol/L RES)and si-PGC-lα+RES+LPS group(si-PGC-lαinterference fragment+hunger medium co-prepared with 10μg/mL LPS and 15μmol/L RES).The results showed that the addition of LPS after si-PGC-lαsignificantly up-regulated the mRNA relative expression levels of contents of the inflammatory factors interleukin-6(IL-6)and interleukin-8(IL-8)(P<0.05 or P<0.01),significantly up-regulated the mRNA relative expression level of the oxidizing factor malondialdehyde(MDA)(P<0.01),significantly up-regulated the mRNA relative expression level of the pro-apoptotic factor B-celllymphoma-2 associated X protein(BAX)and bMECs apoptosis rate(P<0.05 or P<0.01),and significantly down-regulated the mRNA relative expression level of the anti-oxidant gene glutathione peroxidase(GSH-Px)(P<0.01);and RES could reversed this response.When si-PGC-lαwas followed by RES treatment,it failed to alleviate the oxidative stress,inflammatory stress and apoptosis produced by LPS-induced bMECs,suggesting that RES played a role in alleviating LPS-induced inflammatory injury and apoptosis in bMECs by activating the PGC-1α.In conclusion,exogenous addition of RES can alleviate LPS-induced mitochondrial damage,oxidative stress,inflammatory response and inhibit apoptosis of bMECs by activating the PGC-1α.
