摘  要:目的基于网络药理学方法探讨四磨汤口服液在腹部手术后促进肠胃功能恢复的潜在作用机制。方法采用ETCM、TCMSP数据库筛选出四磨汤口服液的活性成分,利用TCMSP靶点预测模型预测活性成分的潜在靶点;利用Genecards、OMIM、Disgenet和DigSee等数据库检索手术后促进肠胃功能恢复的相关靶点;利用Biogenet分别对药物与疾病靶点进行蛋白质相互作用网络构建,并提取交集网络以获得四磨汤治疗胃肠功能紊乱的关键靶点;利用Metascape对关键靶点进行GO和KEGG富集分析。结果得到药效成分37个、药物靶点241个、疾病靶点1315个,最终获得关键靶点24个、KEGG通路144条。四磨汤口服液治疗手术后胃肠功能障碍主要成分为槲皮素、橙皮苷等,关键靶点为ALB、MAPK14、AKT1、MAPK8、EGFR、IGF1、NR3C1、RHOA,关键的生物学进程和通路可能包括PI3K-Akt信号通路、NF-κB信号通路、PPAR信号通路等。结论本研究初步揭示了四磨汤多成分、多靶点、多途径治疗腹部手术后胃肠功能紊乱的机制,为四磨汤的临床合理应用提供了依据。Objective Based on network pharmacology methods,to explore the potential mechanism of Simo Tang oral liquid in promoting postoperative recovery of gastrointestinal function after surgery.Methods Using ETCM and TCMSP databases to screen the active ingredients of Simo Tang Oral Liquid,and using the TCMSP target prediction model to predict the potential targets of the active ingredients;Using Genecards,OMIM,Disgene,and DigSee databases to search for relevant targets that promote postoperative recovery of gastrointestinal function;Using Biogenet to construct protein interaction networks between drug and disease targets,and extracting intersection networks to obtain key targets for the treatment of gastrointestinal dysfunction with Simo Tang;Conduct GO and KEGG enrichment analysis on key targets using Metascape.Results The analysis results obtained 37 pharmacological components,241 drug targets,and 1315 disease targets,ultimately resulting in 24 key targets and 144 KEGG pathways.The main components of Simo Tang oral liquid in treating postoperative gastrointestinal dysfunction include quercetin,hesperidin,etc.The key targets are ALB,MAPK14,AKT1,MAPK8,EGFR,IGF1,NR3C1,RHOA,and the key biological processes and pathways may include PI3K-Akt signaling pathway,NF-κB signaling pathway,PPAR signaling pathway,etc.Conclusions This study preliminarily revealed the mechanism of the multi-component,multi-target,and multi-pathway treatment of gastrointestinal dysfunction after abdominal surgery with Simo Tang,providing a reference for the clinical rational application of Simo Tang.
